Within a eukaryotic organism up to 95 per cent weight for weight of the cell's membranes are used in the formation of intracellular structures such as the nucleus, endoplasmic reticulum ER, golgi, mitochondria, chloroplasts and microbodies, (peroxisomes/glyoxisomes). To allow each membrane bound compartment to carry out its role there is considerable protein traffic between proteins and they must be able to pass across these semi-permeable barriers and in the case of membrane proteins insert into the bilayer. Furthermore the cell must have a means of maintaining proteins at their final functional location against the bulk flow of protein for example in the ER and golgi.

Protein targeting is the study of how newly synthesized proteins reach their ultimate destination. Proteins may be targeted into an intracellular organelle such as the nucleus, mitochondria, or rough endoplasmic reticulum, or they may be secreted or taken up by a cell and distributed tospecific locations such as the endosome or lysosome.
