Book Description
This volume not only discusses various common biobanking topics, it also delves into less-discussed subjects such as what is needed to start a biobank, training of new biobanking personnel, and ethnic representation in biospecimen research. Other chapters in this book span practical topics including: disaster prevention and recovery; information technology; flora and fauna preservation including zoological fluid specimen photography; surgical and autopsy biobanking; biobanking of bodily fluids; biosafety; cutting frozen sections; immunohistochemistry; nucleic acid extraction; and biospecimen shipping. Written in the highly successful Methods in Molecular Biology series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls. Unique and comprehensive, Biobanking: Methods and Protocols is a valuable resource for novice and practicing biobankers, and for end-user researchers. This book aims to bring new insight into the field and expand on current biomedical biobanking studies.

Book Description
Immunohistochemistry and immunocytochemistry are invaluable tools for the visualization of tissue and cellular antigens in diagnostic and biological research environments. The need to obtain accurate, reliable and reproducible results is paramount. It is with this fundamental aim in mind that we have compiled Immunohistochemistry: Essential Methods. We have achieved this by examining each aspect of immunochemistry in turn, with each chapter including detailed information regarding the subject matter in question. Each chapter is written by an expert in their field and includes protocols that are typically used in their own research. Subjects covered are, amongst others, antibodies and their production; selection of reporter labels; immunochemical staining methods and experimental design (both using single and multiple reporter labels); quality assurance; automated immunochemistry; confocal microscopy and electron microscopy. In addition, benefits and limitations of each approach are discussed within the chapters.

Book Description
Since antibodies tagged with markers have been developed, immunocytochemistry has become an important method for identifying tissue substances and the localisation of nucleic acid in tissue by in situ hybridisation in molecular biology. This laboratory book covers the embedding of tissue using less sensitive epoxy resin methods to the more sensitive procedures employing the acrylics. The possibilities are discussed and results are presented so that an understanding of the techniques can be acquired.

Book Description
The most complete, up-to-date reference on antigen retrieval and immunohistochemistry An antigen is a substance that prompts the generation of antibodies and can cause an immune response. The antigen retrieval (AR) technique is in wide use across the globe, and is a critical technique used in medical diagnosis of disease, particularly clinical targeted cancer treatment. Antigen Retrieval Immunohistochemistry Based Research and Diagnostics discusses several scientific approaches to the standardization of quantifiable immunohistochemistry (IHC). Based on the development and application of AR by the editors, this volume summarizes recent achievements in AR-IHC and analyzes numerous cutting-edge issues for future research projects. Featuring contributions from a worldwide group of leading experts and research scientists in the field, this important work: Summarizes the key problems in the four fields of antigen retrieval Discusses the advances of AR techniques and their applications Provides practical methods and protocols in AR-IHC, such as extraction of nucleic acids and proteins for molecular analysis, cell/tissue sample preparation, and standardization and development of various techniques to meet the future needs of¿clinical and research molecular analysis Encourages further research in AR and IHC, particularly how AR methods might be employed for improved test performance and the development of greater reliability and reproducibility of IHC Includes an appendix of related laboratory protocols Antigen Retrieval Immunohistochemistry Based Research and Diagnostics is intended for clinical pathologists, molecular cell biologists, basic research scientists, technicians, and graduate students who undertake tissue/cell morphologic and molecular analysis and wish to use and extend the power of immunohistochemistry. It is also pertinent for most biotechnology companies majoring in development of IHC products. Book Description
Heat-induced antigen retrieval (HIAR) method reported by Shi et al. in 1991 has greatly contributed not only to immunohistochemistry but also to studying gene expressions using archived formalin-fixed and paraffin-embedded (FFPE) specimens. Heating cleaves crosslinks (methylene bridges) in formaldehyde-fixed proteins and extends polypeptides to expose epitopes hidden in the inner portion of antigens or covered by adjacent macromolecules. In this chapter, the following topics are described to reconsider the concept of immunohistochemistry flexibly and to apply HIAR for further immunological studies using a variety of specimens: (1) antigen-antibody interactions in tissues; (2) mechanisms of chemical fixation with formaldehyde, glutaraldehyde, and osmium tetroxide; (3) unmasking of epitopes using HIAR for specimens fixed with chemical fixatives, including highly masked epitopes; (4) a standardized fixative for immunoelectron microscopy-based HIAR; (5) HIAR for conventionally processed electron microscopy specimens embedded in epoxy resins; and (6) effects of antibody diluents on immunohistochemistry.
