"Neuroscience research has been illuminated by the application of light-sensitive proteins, called opsins, in manipulating neuronal function. Optogenetics has been readily applied in mice, however the application of this techniques in non-human primates (NHPs) has produced underwhelming results. These challenges can largely be attributed to the immense size difference between rodent and primate brains. Additional challenges such as adaptive immunity and increased brain complexity may also contribute. We have identified and addressed several challenges that have hindered successful optogenetic manipulations in NHPs including difficulties in selecting an appropriate vector, unreliable viral vector batch quality and efficacy, inefficient injection protocols, tissue damage and light-derived artifacts from poor optrode design, and poor light propagation through brain tissue. This study sough to address these issues through in vivo and in vitro investigations to yield a streamlined methodology for achieving NHP optogenetics. To determine an optimal vector for NHP transgene delivery, we compared the transduction efficiency of a panel of 14 different serotypes of the adeno-associated virus (AAV) on neurons differentiated from Macaca fascicularis embryonic stem cells. AAV6 was ultimately found to produce the highest transduction efficiency, followed by AAV2. We then developed a cohesive methodology for in vivo NHP optogenetics manipulation, including an in-chair viral injection strategy, utilizing opsins possessing red-shifted activation spectrum, and fabricating tapered optical fibers paired with thin electrodes to reduce tissue damage upon insertion. This methodology was then applied in vivo, successfully achieving electrophysiological optogenetic manipulation in the primate lateral geniculate nucleus and electrophysiological and behavioural manipulation in the horizontal and vertical oculomotor centres of the brain stem"--
