This book, which originated in the presentations of a symposium sponsored by the Universidad Nacional Autonoma de Mexico, held in Mexico City on April 14-16, 1980, represents an attempt to analyze some of the most relevant aspects of synaptic transmission. This topic was chosen on the strong belief that the progress of the neurosciences depends to a great extent on the understanding of the basic mechanisms of synaptic function. Rather than selecting only a specific approach or speciality, the book intends to cover this field in a multi disciplinary way. This means that neurochemical, neurophysiological and morphological studies are mingled throughout the book, which hopefully will help the reader to integrate the different faces of the same problem. Across the book the presynaptic component of synaptic transmission and its regulation is stressed much more than the postsynaptic phenomena. Although this might be a limitation, it has the advantage of increasing the focus of the book on a series of events which are gaining importance and interest every day.

Neurons in the nervous system organize into complex networks and their functions are precisely controlled. The most important means for neurons to communicate with each other is transmission through chemical synapses, where the release of neurotransmitters by the presynaptic nerve terminal of one neuron influences the function of a second neuron. Since the discovery of chemical neurotransmission by Otto Loewi in the 1920s, great progress has been made in our understanding of mol- ular mechanisms of neurotransmitter release. The last decade has seen an explosion of knowledge in this field. The aim of Molecular Mechanisms of Neurotransmitter Release is to provide up-to-date, in-depth coverage of essentially all major mole- lar mechanisms of neurotransmitter release. The contributors have made great efforts to write concisely but with sufficient background information, and to use figures/diagrams to present clearly key concepts or experiments. It is hoped that this book may serve as a learning tool for neuroscience students, a solid reference for neuroscientists, and a source of knowledge for people who have a general interest in neuroscience. Any brain activity relies on the interaction of thousands of neurons, each of which integrating signals from thousands of synapses. While neurons are undoubtedly the building blocks of the brain, synapses constitute the main loci of information transfer that lead to the emergence of neuronal code. Investigating synaptic transmission constitutes a multi-faceted challenge that brings together a large number of techniques and expertise ranging from experimental to computational approaches, bringing together paradigms spanning from molecular to neural network level. In this book, we have collected a series of articles that present foundational work aimed at shedding much-needed light on brain information processing, synaptic transmission and neural code formation. Some articles present analyses of regulatory mechanisms underlying neural code formation and its elaboration at the molecular level, while others use computational and modelling approaches to investigate, at synaptic, neuronal and inter-neuronal level, how the different mechanisms involved in information processing interact to generate effects like long-term potentiation (LTP), which constitutes the cellular basis of learning and memory. Antidepressants are generally believed to exert their clinical efficacy by enhancing 5-HT transmission. Interestingly, sustained administration of selective serotonin (5-HT) reuptake inhibitors (SSRIs) strongly suppresses in the first few days the firing activity of 5-HT neurons in the dorsal raphe nucleus (DRN), thereby severely hampering the increase of 5-HT in target regions. Remarkably, the firing activity of 5-HT neurons gradually recovers over the time course of treatment and this recovery is believed to be accounted for by the desensitization of 5-HT1A somatodendritic autoreceptors. Here, we sought to investigate whether additional mechanisms might contribute to the dynamic regulation of excitability of 5-HT neurons during the course of SSRI treatments. Borrowing from the well-described homeostatic strengthening of glutamatergic synapses onto cortical pyramidal neurons following prolonged periods of inactivity, we hypothesized that a similar homeostatic-like regulation of synaptic strength might be operant on 5-HT cells during an SSRI treatment. To test this possibility, we used whole-cell electrophysiological recordings on acute midbrain slices to monitor glutamatergic synapses onto 5-HT neurons. We found that a two-day treatment with the SSRI citalopram induced a robust reduction in both the amplitude and frequency of AMPAR-mediated mEPSCs. We also show that this depression in synaptic strength, induced by an SSRI, is transient since excitatory drive onto 5-HT neurons was enhanced by 7 days of treatments. Altogether, these results document a dynamic regulation of glutamatergic synaptic transmission during the time course of a prolonged treatment with an SSRI. Further elucidation of the cellular and molecular mechanisms driving this synaptic plasticity might identify novel pharmacological target to shorten the delay of antidepressant action.
