Une étape clef pour résoudre la structure de macromolécules biologiques est de déduire la phase des facteurs de structure à partir des intensités diffractées par le cristal. Les méthodes de phasage SAD (Single-wavelength Anomalous Diffraction) et MAD (Multiple-wavelength Anomalous Diffraction) utilisent la variation du facteur de diffusion de quelques atomes, présents dans la structure native ou introduits à dessein, dans un de leurs seuils d'absorption. De fortes variations des facteurs de diffusion anomaux, f' et f", induisent de fortes différences entre les intensités des paires de Friedel mesurées à une ou à plusieurs longueurs d'onde. La phase du facteur de structure est déduite de ces différences. Nous avons apporté de nouveaux développements aux méthodes SAD et MAD à l'aide de dérivés obtenus avec des complexes de lanthanides (f' et f" très élevés). Les premiers complexes étudiés ont été ceux de gadolinium, élément qui présente une raie blanche (f"=28e-) dans son seuil d'absorption LIII ([Lambda]=1.711 Aʿ) et pour lequel la diffusion anomale reste importante (f"=12 e-) avec le rayonnement d'un générateur de laboratoire. Ces complexes permettent de résoudre de novo, par la méthode SAD, la structure de macromolécules qui vont du lysozyme (14 kDa) à partir de données enregistrées au laboratoire, à l'OTCase 3630 (450 kDa par unité asymétrique) à partir de données enregistrées, à l'aide du rayonnement synchroton, au seuil d'aborption LIII du Gd. La fixation dépend des agents de cristallisation : des complexes étudiés se fixent fortement sur des protéines cristallisant avec des sels (lysozyme, catalase et OTCase 3630) et se fixent peu sur des protéines cristallisant avec du PEG 8000 (urate oxydase, [bêta]-lactamase). Cette situation est inversée pour les autres complexes employés. La structure des cinq protéines étudiées a été résolue de novo par la méthode SAD avec ces molécules. Ces complexes de lanthanides présentent un grand intérêt pour la génomique structurale.
