A new class of mutation was induced by X-radiation in Paramecium aurelia . These mutant segregants were called temperature-sensitive mutants ( ts mutants) since they can grow normally at 27° but are inviable at 36°. The maximum yield of ts mutant segregants after 2 kR of X-radiation was about 14% of the segregants viable at 27°. Crosses between persistent ts mutant segregants and wild-type stocks showed that the ts character is inheritable and recessive. Crosses between different ts mutant stocks revealed that these ts loci are widely spread over the genome of Paramecium. Certain subsidiary problems arising in the course of the work and the genetic applications of the ts mutations to genetic analysis of paramecia were discussed.

The purpose of this and future volumes of the Handbook of Genetics is to bring together a collection of relatively short, authoritative essays or annotated compilations of data on topics of~ignificance to geneticists. Many of the essays will deal with various aspects of the biology of certain species selected because they are favorite subjects for genetic investigation in nature or the laboratory. Often there will be an encyclopedic amount o( information available on such a species, with new papers appearing daily. Most of these will be written for specialists in a jargon that is bewildering to a novice and sometimes even to a veteran geneticist working with evolu tionarily distant organisms. For such readers what is needed is a written introduction to the morphology, life cycle, reproductive behavior, and cul ture methods for the species in question. What are its particular ad vantages (and disadvantages) for genetic study, and what have we learned from it? Where are the classic papers, the key bibliographies, and how or mutant strains? A list giving the sym does one get stocks of wild type bolism for unknown mutations is helpful, but it need include only those mutants that have been retained and are thus available for future studies. Other data, such as up-to-date genetic and cytological maps, listings of break points for chromosomal aberrations, mitotic karyotypes, and hap loid DNA values, will be included when available.

This completely revised, updated, and expanded edition has been neces sitated by the many important newer discoveries that have been made since the publication of the first edition. That volume contained almost 2000 references from the 1600s to 1953. Since then and after an extensive search, I have accumulated some 3800 additional titles from the world's literature on Paramecium. After certain titles that largely represented abstracts and preliminary reports that were followed by full research papers were eliminated, approximately 4400 full titles from the 1600s to the present were selected for inclusion in this edition. Most of the titles in the Bibliography are the more recent ones beginning with 1953. I consider the Bibliography an important part of the book: A good title of a paper in a journal is a minature abstract of its contents. Thus, the reader who may be interested in obtaining additional information not found in the book may refer directly to the original source. A cursory examination of the Bibliography will reveal that parame cium research has extended into all branches of biology, including bio chemistry and biophysics. Like other areas of science, paramecium research has become highly specialized and fragmented. I have attempted to organize and present the basic information in one book. Because of space limitations, some items may have been dealt with briefly. It is in such instances that the Bibliography will be found to be invaluable.
