We discuss digital holographic microscopy (DHM), a 3D imaging technique capable of measuring the positions of micron-sized colloidal particles with nanometer precision and sub-millisecond temporal resolution. We use exact electromagnetic scattering solutions to model holograms of multiple colloidal spheres. While the Lorenz-Mie solution for scattering by isolated spheres has previously been used to model digital holograms, we apply for the first time an exact multisphere superposition scattering model that is capable of modeling holograms from spheres that are sufficiently close together to exhibit optical coupling.

Based on the DHPIV technique, two systems were implemented in Laser Flow Diagnostics Lab. My work was to calibrate the systems with different particles, quantify the errors and find the limitations of the two holography systems as a reference for practical use and further development. I also made an attempt to explore the capabilities of the systems for applications. In the first part, calibrations were performed on the shape, position and size accuracy. The work done on the different objects in the four steps of the digital information processing, namely preprocessing, image reconstruction, particle extraction and velocity extraction were discussed, and the lens-based recording was studied. A novel particle extraction method, PECA, which was developed by Gang Pan, was proved out to be more accurate than the conventional PEI method for both depth resolution and size measurement. In the second part, capabilities of the system for applications were demonstrated by the practical tests. (Abstract shortened by UMI.).
