La Résonance Magnétique Nucléaire (RMN) s'est révélée particulièrement efficace dans l'étude des macromolécules biologiques in situ (et in vivo). Les premières applications ont d'abord porté sur l'étude de petites molécules en solution puis, rapidement, l'intérêt s'est déplacé vers l'analyse des protéines en solution. Depuis quelques années, la RMN du solide en rotation à l'angle magique (MAS) est apparue comme une technique adaptée dans l'étude structurale des protéines membranaires dans leurs membranes natives. Il est en effet établi que les deux constituants majeurs des membranes biologiques (lipides et protéines membranaires) sont intimement liés, et l'étude de leurs influences réciproques permettrait d'obtenir une vision plus complète de celles-ci. Afin d'apporter certaines d'informations à cette question, nous avons choisi un modèle particulier : la souche C43(TDE3) et son réseau membranaire formé suite à la sur-expression de la sous-unité b de l'ATP synthase FoFi d'E. co/i. L'étude de la formation de ce réseau, de l'implication de la sous-unité b (RMN MAS 13C/15N) et des lipides (spectrométrie de masse et RMN 31P), a permis d'obtenir certaines données quant à l'importance de ces deux composants dans la mise en place et stabilisation de ce réseau. De plus, au cours de ce projet, l'utilisation combinée de la RMN 'H et du MAS s'est révélée particulièrement adaptée dans l'étude de la dynamique des membranes de cellules entières selon diverses conditions de croissance. De fait, les différentes méthodes développées et utilisées au cours de projet pourraient s'avérer utiles dans l'étude de diverses membranes biologiques, que ce soit du point de vue protéique ou lipidique.
