Abstract: Abstract of thesis entitled Functional Characterization of Liver Intestine-Cadherin (CDH17) in Hepatocellular Carcinoma Submitted by Vivian Wai Man Chan For the degree of Master of Philosophy at the University of Hong Kong December 2006 Liver intestine-cadherin (LI-cadherin, CDH17) is a new cancer biomarker for hepatocellular carcinoma (HCC). However, the molecular mechanism of LI-cadherin on HCC pathogenesis remains unknown. The present study aims to characterize the functions of LI-cadherin and to elucidate the molecular pathway of CDH17 in HCC progression. Gene expression level of CDH17 was determined in HCC cell lines and clinical specimens by TaqMan real time Q-PCR. Protein expressions were determined by western blot and immunohistochemistry. For functional analysis, short hairpin-RNA (shRNA) constructs targeting to CDH17 exon 5 and other exons were stably transfected into HCC cell line of high metastatic potential (MHCC-97H). Alterations of tumor phenotypes were observed and determined by vitro functional assays and in vivo nude mouse model. The present findings indicate that overexpression of LI-cadherin is clinically correlated to the advanced tumor stages (III-IV) and presence of HCC venous infiltration. The metastatic HCC cell lines expressed significantly higher level of CDH17 than the primary HCC counterparts. Functional studies showed that LI-cadherin promoted HCC tumor growth in vitro iand in vivo, and be involved in tumor metastasis by cell adhesion assay and cell invasion assay. Furthermore, LI-cadherin is correlated with the altered expression and localization of β-catenin, cyclin D1 and retinoblastoma (Rb).
