This book intends to be neither a complete survey of the field nor an exhaustive source of references. For these purposes, the use of the extensive compilation "Experimental Immunochemistry" by E. A. KABAT and M. M. MAYER (1962) or the excellent methodological textbook, "Methods in Immunology", by D. H. CAMPBELL, J. S. GARVEY, E. E. CREMER and D. H. SUSSDORF (1963), or the quite comprehensive series "Methods in Immunology and Immunochemistry" by C. A. WILLIAMS and M. W. CHASE (1967) are more suitable. The main purpose of this manual ist to provide students with a simple book which will introduce them to some frequently occurring problems in the three major sections of the immunochemistry of natural products. These are the isolation of the materials, the chemical analysis of the constituents and their structure, and, finally, the assays of the most important biological and immunological activities. In this manual the exercises are simplified and several shortcuts are taken in order to fit them into the framework of a teaching course. The introduction to each exercise gives a brief and elementary explanation of the reaction on which it is based. "Materials and Equipment" lists all tissues or cells, chemicals, glassware, and special equipment which must be available to carry out the exercise, although the very common laboratory tools are usually omitted from the list.

This fourth volume of Contemporary Topics In Immunobiology treats in vertebrate immunity. Specifically, the results represent several approaches to humoral and cellular immunity. It is evident that invertebrates do have function ing immune systems. For example, cellular immunity is characterized by both specificity and memory, but it is still problematical whether vertebrate immune capacity evolved directly from invertebrates. Most of the manuscripts were formally presented at the International Symposium on Invertebrate Pathology, University of Minnesota, August 1972, held in connection with the 25th anniversary celebration of the American Insti tute of Biological Sciences. Human cystic hydatid disease (CHD) caused by the larval stage (hydatid cyst) of the dog tapeworm, E. granulosus, is a major infection with worldwide distribution and variable geographical incidence. The present study evaluates the two most popular techniques, sandwich ELISA and Latex techniques for the diagnosis of human hydatidosis by antigen detection. In the present study, crude protoscolex antigen (CPA) detection was carried out through several steps including the preparation of protoscoleces antigen, production; purification and labeling of rabbit IgG pAb. The Latex technique and ELISA tests have been found to be high sensitive and suitable for the antigen detection of cystic hydatid disease. In conclusion, antigen detection assay is superior and more sensitive than antibody detection assay, especially in diagnosing active infection where hydatid cysts are predominant. Antigen detection assay might be a useful approach for assessment of the efficacy of treatment especially after removal of the cyst. Further studies are recommended to improve the diagnostic efficacy of antigen based ELISA method. Sandwich ELISA and Latex techniques appear to be sufficiently sensitive assays for the detection of human echinococcosis.

Since their original discovery and study at the beginning and middle parts of this century, many investigations have been performed concerning their immunochemistry and physicochemistry, as well as their pharmacologic activities and physiologic effects on the host. It became widely recognized during the beginning of this century that the pyrogenicity of many microbial infections may be associated with endotoxins. Furthermore, some 80 years ago, attempts were begun to "treat" a variety of illnesses including neoplasia, with such "pyrogens", Le. , bacterial endo toxins. Inconclusive results were observed including some detrimental ones as well as, in some cases, beneficial ones. It became widely accepted that during infections with many gram negative organisms the fever occurring in patients, as well as many of the untoward pathophysiological effects of the infections, seemed to be due to the endotoxin the bacteria contained or released. In this regard, septic shock has been studied in detail by many clinicians, physiologists and pharmacologists and attempts have been made to relate the devastating effects of infection on metabolic and physiologic alterations caused by endotoxins. Recently, however, many beneficial effects of endotoxin have also been studied.
