Cryopreservation of fish sperm is a valuable method for restoration of endangered species as well as serving as complementary technique of reproductive manipulation for genetic improvement. In the cryopreservation of sperm, exposure to cryoprotectant prior to freezing is an important factor for many species. Therefore, the present study was conducted to develop an optimal freezing rate in the cryopreservation protocol of red tilapia fish sperm. The results obtained from the present study showed that the main factors that influenced the quality of sperm used in cryopreservation were molarity of glycerol in extender, equilibration time, vapour temperature and individual fish. However, in an attempt to develop an efficient and routine cryopreservation technique of fish sperm, more research needs to be conducted.

Conservation efforts are implemented worldwide in an attempt to mitigate the decline in biodiversity experienced today. As of 2021, over 3000 species of fish worldwide have been classified as critically endangered, endangered, or vulnerable.Primary contributors include climate change, pollution, habitat loss, over fishing, and invasive species. These issues are not being combated fast enough to save species from extinction. Thus, it is vital to develop complementary approaches to conserve biodiversity. An area of research under development is cryopreservation of fish sperm.Sperm cryopreservation provides an excellent method for species conservation by allowing for long-term storage of genetic material without risk of deterioration; therefore,providing a safeguard against species collapse. Unfortunately, cryopreservation is difficult to standardize across fish species due to its physiological complexity as well as biological differences of spermatozoa across species that influence their tolerance to the freeze/thaw process. Thus, species-specific methods must be developed. Blue tilapia(Oreochromis aureus) are of particular interest due to their importance as a food source for humans, excellent adaptability to being in culture, they are easy to breed, reproduce quickly, and tolerate high stocking densities. However, there is limited research on the cryopreservation of blue tilapia sperm, as the focus often lies on Nile and Mozambique tilapia. Therefore, I investigated an optimized cryopreservation protocol for blue tilapia sperm by testing combinations of three different cryoprotectants: dimethyl sulfoxide(DMSO), methanol (MeOH), and glycerol (Gly), with three different slow cooling rates: -1°C/min, -5°C/min, and -10°C/min. After thawing, viability of the protocols was assessed through spermatozoa motility and swimming kinematics as well as cell membrane damage using Trypan Blue membrane stain and a lactate dehydrogenase (LDH) enzyme assay on seminal plasma. Results indicate that DMSO at -5°C/min yielded consistently better outcomes than other treatments, however, due to limited overall success, further investigation is required to achieve the most effective protocol. Finally, percent motilityanalysis and Trypan Blue staining provided the most reliable results while LDH activity and swimming kinematics were less conclusive as viability measures.

The main aim of this study is to determine the effect of the straw volume (0.25 vs. 0.5 mL) on Nile tilapia sperm quality after cryopreservation. Sperm was frozen according to conventional slow freezing procedure and diluted at ratio of 1:3 with ionic extender containing 350 mM glucose and 30 mM Tris containing 10% dimethylacetamide. Diluted semen was equilibrated at 4°C for 10 min and drawn into 0.25-mL or 0.5-mL plastic straws and sealed with polyvinyl alcohol. Samples were frozen 3 cm above of the liquid nitrogen surface and exposed to the liquid nitrogen vapor (≈−140°C) for 10 minutes After this, frozen sperm cells were kept into the liquid nitrogen container (−196°C). The frozen sperm in different volume of straws were thawed in a water bath at 30°C for 20 s (0.25-mL straws) or at 30°C for 30 s (0.5-mL straws), respectively. Fertilization was conducted using 1 ×105 spermatozoa/egg ratio with each straw type. The findings of the present study indicated that cryopreservation of sperm in glucose-Tris-based extender using 0.5-mL straws improved post-thaw progressive motility, duration of progressive motility, and fertilization results (P0.01). On the other hand, differences in term of post-thaw cell viability was not significant among the treatments (P0.01). In conclusion, our results suggest that Nile tilapia sperm can be successfully cryopreserved in Tris-based extenders supplemented with glucose containing 10% dimethylacetamide in 0.5-mL straws.
