Les systèmes d'administration de médicaments (Drug Delivery Systems (DDSs)) sont des formulations ou des dispositifs utilisés pour améliorer les performances de médicaments peu efficaces. Les Metal-Organic Frameworks (MOFs) de taille nanométrique sont des vecteurs de médicaments potentiellement intéressant de part leur capacité à incorporer de grandes quantités de principes actifs dans leurs pores, et dont la surface peut être modifiée, leur donnant une meilleure stabilité et efficacité. L'analyse de la structure d'un DDS est une étape clé pour guider les chimistes vers la synthèses de matériaux toujours plus performants. La spectroscopie RMN à l'état solide est une technique idéale pour l'analyse de ces architectures supramoléculaire car elle fourni des informations sur la localisation du médicament, l'interaction médicament-MOF et la structure du MOF, ainsi que sur le processus de dégradation, qui conduit à la libération du médicament. Dans cette thèse, nous avons exploré les potentialités de la RMN du solide associée, quand nécessaire, au marquage isotopique pour la caractérisation poussée de DDSs à base de MOFs. Une attention particulière a été portée sur l'utilisation des hétéronoyaux (19F, 27Al, 31P, 13C, 17O) présent dans le principe actif ou dans le nanovecteur et qui, au prix d'une sévère perte de sensibilité, offre beaucoup plus d'information que le noyau 1H.

TWENTY-FIVE years ago in Russia, Zavoisky made the first experimen tal observation of electron spin resonances; and a year later Purcell, Torrey and Pound at Harvard and Bloch, Hansen and Packard at Stanford ('The Harvard of the West') observed nuclear magnetic resonances for the first time. In both cases (ESR and NMR) the phenomena had been previously predicted, and Gorter - surely one of the most unlucky experimenters of our time - had made his noble attempts to detect magnetic resonances. Purcell et aZ. , having beaten their radar swords into scientific ploughshares, used a re 3 sonant coaxial cavity, filled the inductive part with 850 cm of paraffin, and produced a resonance with a signal-to-noise ratio of 20. They predicted that the sensitivity could be increased several hundred-fold and foresaw applications in determining magnetic mo ments, investigating spin-lattice coupling and measuring magnetic fields. Their letter reached the editor of Phys. Rev. on Christmas Eve 1945, and the basis of NMR in the solid state was laid. Bloch et aZ. , fortunately working independently, established the entirely different, crossed-coil approach to the observation of NMR in a water sample, which they doped with paramagnetic ions to reduce the relaxation times. They had laid the foundation of liquid-state NMR. Their letter to Phys. Rev. was received on 29 January 1946.

Why to apply solid-state NMR? - By now, we should have learned that NMR is mainly used for the study of molecules in solution, while x-ray diffraction is the method of choice for solids. Based on this fact, the two recent 'NMR-Nobelprizes' went indeed into the liquid phase: my own one eleven years ago, and particularly the most recent one to Kurt Wuthrich. His prize is beyond any doubts very well justified. His contribution towards the study of biomolecules in solution, in their native (or almost native) environment is truly monumental. We all will profit from it indirectly when one of our future diseases will be cured with better drugs, based on the insightful knowledge gained through liquid-state NMR. Two fields of NMR are still left out of the Nobel Prize game: magnetic reso nance imaging (MRI) and solid-state NMR. The disrespect for MRI in Stockholm is particularly difficult to understand; but this is not a subject to be discussed at the present place. Solid-state NMR is the third of the three great fields of NMR, powerful already today and very promising for the near future.

The field of Nuclear Magnetic Resonance (NMR) has developed at a fascinating pace during the last decade. It always has been an extremely valuable tool to the organic chemist by supplying molecular "finger print" spectra at the atomic level. Unfortunately the high resolution achievable in liquid solutions could not be obtained in solids and physicists and physical chemists had to live with unresolved lines open to a wealth of curve fitting procedures and a vast amount of speculations. High resolution NMR in solids seemed to be a paradoxon. Broad structure less lines are usually encountered when dealing with NMR in solids. Only with the recent advent of mUltiple pulse, magic angle, cross-polarization, two-dimen sional and multiple-quantum spectroscopy and other techniques during the last decade it became possible to resolve finer details of nuclear spin interactions in solids. I have felt that graduate students, researchers and others beginning to get involved with these techniques needed a book which treats the principles, theo retical foundations and applications of these rather sophisticated experimental techniques. Therefore I wrote a monograph on the subject in 1976. Very soon new ideas led to the developement of "two-dimensional spectroscopy" and "multiple-quantum spectroscopy", topics which were not covered in the first edition of my book. Moreover an exponential growth of literature appeared in this area of research leaving the beginner in an awkward situation of tracing back from a current article to the roots of the experiment.

Manipulation and Dilution Tools for Ruling Abundant Species "NMR is dead" was the slogan heard in the late 1960s at least among physicists, until John S. Waugh and his co-workers initiated a series of new NMR experiments, which employed the coherent modulation of interactions by strong radiofrequency fields. A wealth of new phenomena was observed, which are summarized in the introduction for the convenience of the unbiased reader, whereas Section 2 collects the basic spin interactions observed in solids. Line-narrowing effects in dipolar coupled solids by the application of multiple pulse experiments are extensively discussed in Section 3. Numerous extensions of the basic Waugh, Huber, and Haeberlen experiment have been developed by different groups and have been applied to the nuclei IH, 9Be, 19F, 27Al, 31p, 63CU in solids. Application of this technique to a variety of systems is still in progress and should reveal interesting insights into weak spin interactions in solids. It was soon realized that rare spins could be used as monitors for molecular fields in the solid state; however, rare spin observation is difficult because of the small signal-to-noise ratio. Pines, Gibby, and Waugh introduced a new concept of cross-polarization, based on ideas of Hahn and co-workers, which allows the detection ofrare spins with increased sensitivity. The dynamics involved are treated in detail. Other sections merely list results obtained by the techniques described and demonstrate their usefulness in the investigation of dynamical problems in molec ular and solid state physics.
