Advances in the field of glycoprotein hormones necessitated a second international symposium on this topic, held March 11 to 14, 1993, in Santa Barbara, California, and again sponsored by Serono Symposia, USA. The meeting was twofold in its concept: (1) the dissemination of the current research in the field and (2) honoring three scientists who have greatly contributed to this field, Drs. Harold Papkoff, Robert Ryan, and Darrell Ward, upon their retirements. We were honored to have Dr. John Pierre present as a participant at the meeting and also serving as master of ceremonies at the banquet. Certainly one of the highlights at the first meeting, held in 1989, was the cloning of the CG/LH receptor. This second meeting was also filled with insightful and innovative scientific presentations. Significant advances in the regulation of gene transcription of the gonadotropins were presented by Pamela Mellon, James Hoeffler, John Nilson, Bruce Weintraub, and Joel Habener. These presentations reflected major advances from the in the understanding concepts offered at the last meeting. Other advances of hormone structure/function included the continuing work by Joyce Lustbader on the determination of the three-dimensional structure of hCG, including a new approach using nonisotopic labels for 3-D and 4-D NMR imaging. New insights into biosynthesis of hCG were proposed by Irving Boime. Studies directed at identifying receptor contact regions were presented by Henry Keutmann and David Puett.

The peptide hormones are small proteins that regulate cellular metabolism through their specific interactions with tissues of the endocrine, nervous, and immune systems, as well as in embry onic development. During the past ten years, refinements in the techniques of recombinant DNA technology have resulted in the cloning of genes encoding approximately 50 different hormonal and regulatory peptides, including those in which the peptides themselves and the mRNAs encoding the peptides are present in only trace amounts in the tissues of origin. In addition to provid ing the coding sequences of recognized hormonal and regulatory peptides, gene sequencing has uncovered new bioactive peptides encoded in the precursor pro hormones that are then liberated along with the hormonal peptides during cellular cleavages of the precursors. The encoding of multiple peptides in a single mono cistronic mRNA appears to be a genetic mechanism for the gener ation of biologic diversification without requiring amplification of gene sequences. Two of the objectives in the assembly of this book are to pre sent, in one volume, the known primary structures of the genes encoding several of the polypeptide hormones and related regulatory peptides, and to provide an account of the various ap proaches that have been used to identify and select the cloned genes encoding these polypeptides. The contents of the two in troductory chapters are intended to provide the reader with a brief background of the approaches to gene cloning and the struc ture and expression of hormone-encoding genes.

In September of 1977 scientists from many countries met at the Asticou Inn in Maine to present and discuss papers written especially for this monograph. The presentations were informal and directed to the special interests of the audience in order to generate discussions. The authors, many of whom are pioneers and leaders in their field, then had the oppor tunity to revise their contributions, which were brought together with the edited discussions to form this volume. The basic research studies presented here are important because of the essential role of gonadotropins in regulating the ovary and testis. This monograph will therefore be of interest to those concerned with fertility regulation, population control, possible new methods for contraception, and to those concerned with reproduction in domestic animals. Re searchers in other fields may find this monograph useful, as it has been de termined that gonadotropins are secreted by many tumors and are im plicated in many cancers. Human choriogonadotropin also seems to be found in most, if not all, cells of the human body. The significance of this, however, is unknown.
