Dentinogenesis is a complex process of dentin formation. It involves differentiation of odontoblasts, the cells that make dentin, and the formation of a unique extracellular matrix, known as dentin. Dentin is ultimately produced through the interaction of collagen fibers, non-collagenous proteins and hydroxyapatite (HA) crystals. Odontoblasts are cranial neural crest (CNC) ectomesenchymal derived cells. Trps1 is a novel transcription factor believed to play an important role in both tooth formation and the mineralization process. TRPS1 gene mutations are known to result in dental and skeletal abnormalities manifested in humans as tricho-rhino-phalangeal syndrome (TRPS) and Ambras syndrome. Trps1 expression varies throughout odontoblast development. Strongly expressed in preodontoblasts, Trps1 then decreases in mature secretory cells at onset of mineralization; thus its downregulation coincides with terminal differentiation of odontoblasts. Our laboratory recently demonstrated that sustained high expression of Trps1 in secretory odontoblasts represses secretion and mineralization of dentin matrix in vitro. However, the specific role Trps1 plays in dentinogenesis remains unclear. To better understand the function of Trps1 in odontoblast maturation, ex vivo tooth organ culture studies were performed. Trps1 knockout mice (KO) are perinatal lethal, therefore dentinogenesis has not been studied in these mice until now. Analyses were performed to define whether Trps1-deficiency affects odontoblast maturation and/or odontoblast function. We found that Trps1 KO mice were able to form differentiated odontoblasts, capable of secreting a predentin matrix. However, mature odontoblasts in Trps1 KO mice were unable to produce matrix vesicles, an important component that supports the mineralization process. Altered expression of several key osteogenic factors (Runx2, Sp7, TNAP) in Trps1 KO tooth organs revealed that Trps1 functions in both a cell autonomous and non-autonomous fashion in different areas of the developing molar. Furthermore, analyses of embryonic day 18.5 Trps1 KO molars revealed Trps1-deficienct mice exhibit a smaller first mandibular molar compared to WT, mimicking a dental phenotype seen in human patients affected with TRPS. Results of this project have provided first insight into the role of Trps1 in odontoblast maturation and allowed for better understanding of the molecular mechanisms of dentin formation.

Sequential and reciprocal interactions between oral epithelial and cranial neural crest-derived mesenchymal cells give rise to the teeth and periodontium. Teeth are vital organs containing a rich number of blood vessels and nerve fibers within the dental pulp and periodontium. Teeth are composed by unique and specific collagenous (dentin, fibrillar cementum) and non-collagenous (enamel) highly mineralized extracellular matrices. Alveolar bone is another collagenous hard tissue that supports tooth stability and function through its close interaction with the periodontal ligament. Dental hard tissues are often damaged after infection or traumatic injuries that lead to the partial or complete destruction of the functional dental and supportive tissues. Well-established protocols are routinely used in dental clinics for the restoration or replacement of the damaged tooth and alveolar bone areas. Recent progress in the fields of cell biology, tissue engineering, and nanotechnology offers promising opportunities to repair damaged or missing dental tissues. Indeed, pulp and periodontal tissue regeneration is progressing rapidly with the application of stem cells, biodegradable scaffolds, and growth factors. Furthermore, methods that enable partial dental hard tissue repair and regeneration are being evaluated with variable degrees of success. However, these cell-based therapies are still incipient and many issues need to be addressed before any clinical application. Odontoblast differentiation is dependent on multiple signaling molecules, including growth factors stored in the extracellular dentin matrix. Our previous studies have demonstrated that Fibroblast Growth Factor 2 (FGF2) exerted stage-specific effects on odontoblast differentiation in primary dental pulp cultures. FGF2 stimulated differentiation of functional odontoblasts expressing high levels of Dmp1 from early progenitors but inhibited the terminal differentiation of functional odontoblasts to fully differentiated odontoblasts expressing high levels of Dspp. These stimulatory effects involved activation of FGFR and ERK1/2, and increased levels of expression of Bmp2, Runx2, and Osx (components of BMP signaling) suggesting the involvement of BMP signaling in FGF2-induced Dspp expression. Therefore, the overall goal of the studies outlined in this thesis was to examine the role of BMP signaling in stimulatory effects of FGF signaling on odontoblast differentiation of dental pulp cells. We showed that BMP2 did not affect the extent of mineralization but rapidly (within ~12-24 hrs) stimulated expression of Dspp and intensity of DSPP-Cerulean transgene in a concentration-dependent manner without affecting the percentage of DSPP-Cerulean+ odontoblasts. In contrast to Dspp, BMP2 rapidly (within ~24-48 hrs) decreased expression of Dmp1, Bsp, DMP1-mCherry, and BSP-GFP in a concentration-dependent manner. Inhibition of the BMP and FGF signaling pathways by noggin and SU5402, respectively, did not have marked effects on the extent of mineralization but decreased expression of Dspp stimulated by these growth factors. These inhibitory effects were long-lasting and observed up to 14 days after removal of the growth factors. Additional experiments in primary bone marrow stromal cells cultures demonstrated that early and limited exposure to BMP2 did not affect mineralization but decreased expression of Dmp1, Bsp, and respective transgenes. Taken together, our results demonstrated that activation of the canonical BMP signaling pathway during the proliferation phase of in vitro growth resulted in significant increases in the expression of Dspp, and these increases were mediated via reciprocal interaction of the BMP and FGF signaling pathways. These data will further elucidate the perspectives of using both FGF2 and BMP2 in dentin regeneration applications.
