Particulate solids such as pharmaceutical powders often require size enlargement processes to improve the manufacturing properties like flowability. For that reason, dry granulation by roll compaction has been widely used in the pharmaceutical industry. The process consists of compressing powders between two counter-rotating rolls to produce ribbons that will be subsequently milled into granules. The obtained granules are tableted for oral dosage. In this process there are two main limitations: the existence of different designs of the roll compactors, milling systems and the interaction between process parameters and raw material properties are still a challenge and the roll-compaction process leads to an inferior tensile strength of tablets compared with direct compression. These aspects are investigated in this work. In the first part of this thesis, an analysis on the effect of different roll-compaction conditions and milling process parameters on ribbons, granules and tablet properties was performed, highlighting the role of the sealing system and the ribbon density distribution characteristics. In the second part, die compaction of roll-compacted powders, as the last stage of the process, is further investigated in terms of experimental analysis (effect of the granule size and composition and stress transmission measurements) and modelling the compaction behavior of granules.

Dry granulation / roller compaction is an enlargement process by agglomeration of the primary powders without adding any solvent. This operation makes it possible to homogenize the properties of pharmaceutical formulations and improve the flow properties necessary for the manufacture of tablets using rotary presses. In this process, temperature variations in the product are rarely considered. However, recent studies have shown that for pharmaceutical powders, such variations can lead to changes in mechanical and physico-chemical properties. In this thesis, a new method based on in-line measurements of powders temperature compacted by a roller press is developed using an infrared camera. Temperature distributions on the surface of the compact were determined using infrared radiation emitted by the compact after leaving the gap. These distributions showed a correlation with the microstructural properties of the compact, confirming the potential of this procedure in monitoring product quality. To understand the potential sources of the heating of pharmaceutical products in dry granulation, a heat balance model was established in anticipation of the numerical simulation of the thermomechanical coupling of the process by finite element method. As the thermal properties of the compact depend on the relative density, the thermal conductivity and the specific heat were characterized for two model pharmaceutical excipients. In addition, the impact of dry granulation on the rearrangement and densification of granules in die compression is studied using a mathematical model (Cooper-Eaton) which allowed to a better understanding of the contribution of dry granulation to tableting process.

Granulation is the process by which a large number of fine particles (primary particles) are agglomerated together in the presence of a liquid to form a smaller number of larger particles (granules). The process of granulation can be carried out as dry or wet depending upon process conditions. Dry granulation is where the powdered particles, with or without the binder, are compressed together under high pressure to form granules. Wet granulation begins with the addition of a liquid binder to the powders that are to be granulated. There are well known and established techniques such as high shear mixers and fluidised beds which are currently used for the wet granulation of materials. Being batch processes these techniques can process material of variable quality and consistency. In the pharmaceutical industry the production of granules is still mostly based on a batch concept. Continuous process forms are considered to be feasible for only large volume productions and too inflexible for product changes which can occur daily. However, it is gaining in popularity recently since it provides several advantages over batch processes. Techniques such as roller compaction and spray drying are used for the continuous production of granules. In this context, twin-screw granulation can be seen as the latest process form for continuous wet granulation. Some studies have reported the utility of this equipment in continuous wet granulation. However, there are many other possibilities still left undiscovered. The present study was targeted to characterize the process of wet granulation in a twin screw granulator. The effect of variables like powder feed rate, liquid to solid ratio, screw speed, viscosity of granulation liquid on the granulation behavior and granular properties were studied. To determine the effect that each of these had on the properties of the granules, analysis was carried out to determine the size, shape, flowability and strength of the granules. The flow of powder in the twin screw granulator was studied using PIV. Finally, the granulation process along the length of screw was studied. The data will be useful to map the process of granulation using a twin screw granulator.

This book had its origins in a meeting between two (relatively) young particle technology researchers on Rehobeth Beach in Delaware in 1992 near the holiday house of Reg Davies (then Director of the Particle Science and Technology Research Center in Dupont). As we played in the sand, we shared an excitement for developments in particle technology, especially particle characterization, that would lead operations such as granulation to be placed on a sound scientific and engineering footing. The immediate outcome from this interaction was the development of new industry short courses in granulation and related topics which we taught together both in Australia and North America. This book follows closely the structure and approaches developed in these courses, particularly the emphasis on particle design in granulation, where the impact of both formulation properties and process variables on product attributes needs to be understood and quantified. The book has been a long time in the making. We have been actively preparing the book for at least five years. Although the chapters have relatively good bibliographies, this book is not a review of the field. Rather it is an attempt by the authors to present a comprehensive engineering approach to granulator design, scale up and operation. It is exciting for us to see the explosion of research interest around the world in this area in the last five to seven years. Some of the most recent work will have to find its way into the second edition.

In the pharmaceutical industry, dry granulation by roll compaction is a process of size enlargement of powder into granules with good flowability for subsequent die compaction process. Understanding the roll compaction process and optimizing manufacturing efficiency is limited using the experimental approach due to the high cost of powder, time-consuming and the complexity of the process. In this work, a 3D Finite Element Method (FEM) model was developed to identify the critical material properties, roll press designs and process parameters controlling the quality of the product. The Drucker-Prager Cap (DPC) model was used to describe the powder compaction behavior and was determined based on standard calibration method. To overcome the complexity involving two different mechanisms of powder feeding by the screw and powder compaction between rolls, a novel combined approach of Discrete Element Method (DEM), used to predict the granular material flow in the feed zone and the Finite Elements Method (FEM) employed for roll compaction, was developed. Lastly, for a more realistic roll compaction modelling, allowing the fluctuation of the gap between rolls, a Coupled-Eulerian Lagrangian (CEL) approach was developed. FEM simulation results clearly show the effect of different process parameters on roll pressure and density distribution in the compaction zone of powder between the rolls. Moreover, results show that using a cheek-plates sealing system causes a nonuniform roll pressure and density distribution with the highest values in the middle and the lowest at the edges. On the other hand, the resultant pressure and density distributions with the rimmed-roll obtained higher values in the edges than in the middle and overall a more uniform distribution. The combined DEM-FEM methodology clearly shows a direct correlation between the particle velocity driven by the screw conveyor to the feed zone and the roll pressure, both oscillating in the same period. This translates into an anisotropic ribbon with a density profile varying sinusoidally along its length. To validate the results, the simulations are compared with literature and experimentally measured values in order to assess the ability of the model to predict the properties of the produced ribbons.
