This aim of this work is to develop ultrasensitive electrochemical biosensors with high affinity toward ochratoxine (OTA) and aflatoxine M1 (AFM1). In order to obtain the best analytical performances, we associated nano-materials in the transducer construction: conducting polypyrrole polymer and poly(amido-amine) dendrimères. Thanks to this association, we benefited from the conducting material's electrical properties, and the large active detection surface dendrimers. For the bimolecular sensing part, we used specific DNA aptamers which allowed us to quantify mycotoxines at nM concentrations. In addition, the different aptamer based biosensors present a very large dynamic ranges. We also demonstrated through the use of different sizes of dendrimers, that the sensitivity depend not only in the affinity between bioreceptors and their target molecules, but also in the physico-chemical properties of the biosensor.

The technique of "systematic evolution of ligands by exponential enrichment" has transformed the premise of biorecognition to encompass nucleic acids in addition to antibodies.These synthetic counterparts - aptamers - are DNA or RNA strands that have been selected through iterative rounds of in vitro selection to bind with affinity to analytes (from small molecules to proteins) of choice. In this thesis, the voltammetric aptamer-based detection of lysozyme was first explored based on charge density changes on aptamer-modified electrodes upon analyte binding. Different optical and electrochemical methods were then tested for the detection of ubiquitous glycanhydrolaselysozyme was first explored based on charge density changes on aptamer-modified electrodes upon analyte binding. Different detection methodologies (optical and electrochemical methods) were then tested for the detection of MUC1(mucin 1),a glycoprotein overexpressed in elevated concentrations in the serum of patients with epithelial adenocarcinomas; this augments the importance for MUC1 detection as an alternative for ealry cancer diagnostics. Optical biosensors developed for the MUC1 detection was through a DNA hybridization scheme with quantum-dot based flouorescence readout. Electrochemical methods were based on either direct attachment of a redox center, methylene blue(MB), to the aptamer strands,or adding free MB to the electrolyte solution(intercalative binding). In both cases, quantitative detection of MUC1 was possible, although direct labeling showed a better sensitivity. To further improve the sensitivity, an intricate biochemical assay was explored featuring the fusion of anti-thrombin and anti-MUC1 aptamers, in the hope of generating a pico-Molar assay for MUC1 via the MUC1- dependent allosteric upregulation of the proteolytic activity of human a-thrombin.
