Since cytotoxic drugs were first developed, their damaging effects on bone marrow have attracted considerable interest. We now know that the bone marrow is an organ with rapid cell renewal in which maintenance of the steady-state depends on a high rate of cell production, and that the bone marrow toxicity of cytotoxic agents can be explained by the perturbation of such steady-state conditions. Experimental researchers have analysed in detail the effects of cytotoxic agents upon defined hematopoietic cell populations to define the mechanism of action and the location within the cell cycle at which cytotoxic drugs exert their cytocidal action. From such studies, a classification of cytotoxic agents according to their action upon cells during different phases of the cell cycle has been proposed [80-82]. Others have used the acute perturbation of the hematopoietic systems, caused by application of single high doses of cytotoxic agents, to analyse the patterns of depletion and reconstitution of the various hematopoietic compartments in an effort to shed light on the regulatory mechanisms that are responsible for the delicately balanced cell production of the hematopoietic cell renewal systems. A great deal of information on the structure of the hematopoietic systems has come from such studies, and kinetic parameters of these systems have been defined. To clinicians, the hematopoietic toxicity of antineoplastic agents has been an undesired though unavoidable side effect of almost all cytotoxic drugs introduced into clinical practice.

Research on aplastic anaemia has until recently been limited to clinical description, morphology and epidemiology. New methods to culture haemopoietic cells, and advances in our knowledge of proliferation and differentiation in the haemopoietic cell system .opened a new area of scientific interest for this "prototype" of haemopoietic failure. In addition, bone marrow transplantation became not only a clinical method of treatment, but also a source of data useful for the discussion of pathophysiological models of aplastic anaemia. This situation prompted us to arrange an international con ference on aplastic anaemia, with particular emphasis on its patho physiology and the rationals of the current therapeutic approaches. This conference was held at Schloss Reisensburg from July 20-22, 1978 with the participation of both experimental and clinical scientists active in this field or in related areas of research. The proceedings of the symposion reflect the present knowledge as well as the many new questions which arose from the discussions. Although an annual review in any field of scientific investigation can never be as current as desired, especially in the cell cycle field, we hope that this volume will be helpful to students, to recent graduates considering a de1liation in subject and to investigators at the fringe of the cell cycle field wishing to bridge frontiers. An instructive approach to many subjects in biology is often to make comparisons between evolutionary distant organisms. If one is willing to accept that yeast represent a model primitive eukaryote, then it is possible to make some interesting comparisons of cell cycle control mechanisms between mammals and our little unicellular cousins. By and large unicellular organisms have no need for intracellular communication. With the exception of the mating phenomenon in S. cerevisiae and perhaps some nutritional sensing mechanisms, cellular division of yeast proceeds with complete disregard for neighbourly communication. Multicellular organisms on the other hand, depend entirely on intracellular communication to maintain structural integrity. Consequently, elaborate networks have evolved to either prevent or promote appropriate cell division in multicellular organisms. Yet, as described in chapter two the rudimentary mechanisms for fine tuning the cell division cycle in higher eukaryotes are already apparent in yeast.
