L’objectif de ce travail est d’améliorer l’analyse de composés pharmaceutiques, réputée difficile. De nouvelles techniques, la chromatographie en phase liquide à haute température (HTLC) et à haute pression (UPLC) sont utilisées. Ces techniques présentent de nombreux avantages. La très haute pression (1000 bars) permet d’obtenir des séparations ultrarapides. La haute température peut également permettre de diminuer les temps d'analyse. Elle conduit, par ailleurs, à l’amélioration de la forme des pics des composés basiques, à une augmentation de leur efficacité et à des modifications de sélectivité. Ces avantages sont illustrés par des applications de séparations de médicaments. Des systèmes chromatographiques, définis par une phase stationnaire, une phase mobile et une température, sont comparés entre eux en terme d’asymétrie, de sélectivité et de capacité de pics.
