Book Description
There is now considerable genetic evidence that the type 4 allele of the apolipoprotein E gene is a major susceptibility factor associated with late-onset Alzheimer's disease, the common form of the disease defined as starting after sixty years of age. The role of apolipoprotein E in normal brain metabolism and in the pathogenesis of Alzheimer's disease are new and exciting avenues of research. This book, written by the most outstanding scientists in this new filed, is the first presentation of results concerning the implications of apolipoprotein E on the genetics, cell biology, neuropathology, biochemistry, and therapeutic management of Alzheimer's disease.

Book Description
This volume contains the proceedings of the 14th Colloque Médecine et Recherche of the Fondation Ipsen pour la Recherche Thérapeutique devoted to Alzheimer's Disease and dedicated to the epidemiological study of this dementia, a very important issue because the incidence and prevalence of Alzheimer's Disease rise exponentially with age. Epidemiological findings not only confirm dementia as a major challenge for the coming years but also contribute defining risk factors, predicting and may be preventing this disease.

Book Description
This volume addresses what is currently known about the use of ApoE genotyping in diagnosing and predicting Alzheimer's disease. Because ApoE genetic testing has such far-reaching consequences, this volume will be of use not only to those in the medical and scientific community involved in the care of patients with Alzheimer's disease, but also to those concerned with the ethical, legal, epidemiological and public health policy issues as well. A summary will also be included of the results of working groups defining the issues, asking methodological questions, and discussing issues concerning genetic counselling and research-related questions.

Book Description
The mathematical model presented in this book, based on partial differential equations (PDEs) describing attractant-repellent chemotaxis, is offered for a quantitative analysis of neurodegenerative disease (ND), e.g., Alzheimer's disease (AD). The model is a representation of basic phenomena (mechanisms) for diffusive transport and biochemical kinetics that provides the spatiotemporal distribution of components which could explain the evolution of ND, and is offered with the intended purpose of providing a small step toward the understanding, and possible treatment of ND.

Book Description
The aggregation of amyloid-beta by self-assembly into oligomers or amyloids is a central event in Alzheimer’s disease. In this dissertation, I propose a mathematical model as a set of coupled kinetic equations that governs the self-assembly of amyloid-beta filaments in the presence of transition-metal ions. Metal ions have been hypothesized as an important factor in the pathogenesis of AD. There is a considerable literature supporting the impact of metal ions such as copper (Cu), zinc (Zn) and iron (Fe) in many critical aspects of AD and other neurodegenerative diseases. Our study includes Cu and Zn as main transition-metal ions, where their coordination to amyloid-beta regulates the aggregation process in vivo. Metal ions mostly affect the nucleation phase and change both the structure and the charge of amyloid-beta. Our model describes the general features of the kinetics of fragmenting filamentous structures. The numerical simulations reveal a four timescale dynamics related to three important events, which are the formation of the amyloid-metal, the homogeneous aggregation of the filaments and the non-homogeneous aggregation of the protein-metal. The method of singular perturbation is used to discern between these timescales. These results are studied in the framework of slow-fast systems. We also compare the metal with the non-metal dynamics and apply optimization theory for realistic values of reaction rates. Simulations shows that in certain cases the presence of metal accelerates the aggregation of filaments in a drastic way.
