The polydentate, water-soluble bisphosphines $\rm (HOH\sb2C)P(CH\sb2)\sb{X}S(CH\sb2)\sb{Y}S(CH\sb2)\sb{X}P(CH \sb2OH)\sb2$ (X = 2, Y = 3, 3; X = 2, Y = 4, 9; X = 3, Y = 3, 12), $\rm (HOH\sb2C)P(CH\sb2)\sb2SC\sb6H\sb4S(CH\sb2)\sb2P(CH\sb2OH)\sb2$ (6), $\rm(HOH\sb2C)P(CH\sb2)\sb2P(C\sb6H\sb5)(CH\sb2)\sb2P(CH\sb2OH)\sb2$ (15), and $\rm(HOH\sb2C)P(CH\sb2)\sb2(CH\sb3)N (CH\sb2)\sb2N(CH\sb3)(CH\sb2)\sb2P(CH\sb2OH)\sb2$ (18), were synthesized in near quantitative yields by formylation of the appropriate phosphine hydrides in the presence of formaldehyde in oxygen-free ethanol. The reactions of 3 and 6, in aqueous solution, with (Pt(COD)) (Cl)$\sb2$ and $\rm\lbrack Pd(C\sb6H\sb5CN)\sb2\rbrack (Cl)\sb2,$ in methylene chloride, produced the water-soluble Pt(II)- and Pd(II)-complexes 19-22 in near quantitative yields. The reactions of 3, 9, and 12 with $\rm \lbrack ReO\sb2(C\sb5H\sb5N)\sb4\rbrack (Cl), $ in refluxing water, provided the Re(V)-complexes 23-25 in excellent yields. The reactions of 15, in aqueous solution, with $\rm\lbrack Pt(COD)\rbrack (Cl)\sb2,\ \lbrack Pd(C\sb6H\sb5CN)\sb2\rbrack (Cl)\sb2,\ \lbrack RhCl(1,5$-$\rm C\sb8H\sb{12})\rbrack\sb2,$ or $\rm\lbrack ReO\sb2\rbrack(PPh\sb3)\sb2\rbrack, $ in methylene chloride, provided for metallic complexes 26-30 in near quantitative yield. X-ray crystallographic analysis of compounds 20, 23, 24, and 25 established the molecular constitution of the metallic complexes. All of the compounds were characterized by $\sp1$H, $\sp{13}$C and $\sp{31}$p NMR spectroscopy, FABMS, and C and H analysis. Radiochemical in vitro investigations of 3 and 12 with technetium-99m provided evidence for the formation of complexes that were stable over a wide range of pH in excess of 24 hours. In vivo biodistribution analyses of the $\rm\sp{99m}Tc$-complexes in Sprague-Dawley rats provided for evidence of efficient clearance of the complexes at 2 hours post-injection via either the renal-urinary or hepatobiliary pathways. The results of these studies demonstrate the potential utility of thioether-functionalized, water-soluble, bisphosphines as bifunctional chelating agents for the labeling of specific biomolecules in nuclear medicinal applications.
