This volume deals mainly with morphologic, ontogenic and metabolic aspects of phagocytosis and factors affecting RES function. The first section contains chapters on the morphology and surface properties of macrophages. The second section deals with the ontogeny and development of B and T lymphocytes as well as macrophages. The third section covers biochemical and physiologic parameters of mononuclear cells and of polymorphonuclear leukocytes, e.g., cyclic nucleotide and iron metabolism, chemotaxis and enzymatic activity, while the fourth is concerned with the phagocytic event. The fifth section relates to factors affecting RES function with discussions on surface receptors of B lymphocyte activation by antigens in regard to the effect of ligand concentration and induction time, lipopolysaccharide effects in vivo and in vitro as related to RES cells including a correlation of lipopolysaccharide structure with biological activity and, finally, the role of the RES in shock and trauma. Summaries of two workshops are also contained in this volume, the first workshop was concerned with the biochemistry and physiology of the RES; the second dealt with pharmacologically important aspects of RES metabolism and function. It is anticipated that the presentation of symposia and scientific papers and the workshop summaries will reflect the intense activity that is taking place in this multidisciplinary field. A better understanding of the Reticuloendothelial System is directly relevant to both conceptional and practical aspects of not only immunology, but other areas of medical and biological sciences in general.

An extraordinary development of the knowledge, concepts and biomedical applications has occurred during the past two decades in the biological sciences, including the Reticuloendothelial Sys tem (RES). For example, it is now widely recognized that distinct classes of cells are involved in the recognition of "exogenous" invaders of the body such as microorganisms and other foreign an tigens, as well as of "endogenous" parasites represented by trans formed neoplastic cells and altered "self" antigens. Prominent among cell populations involved in such recognition of antigens and subsequent immune responses are those constituting the RES. In recent years, there has been much discussion and indeed contro versy as to what constitutes such a system and even whether the term "RES" is appropriate. Some investigators feel that the phago cytes are the most important cells of the RES as they playa major role in the defense mechanism of the host. Mononuclear phagocytes include tissue macrophages ~s well as circulating monocytes and their precursors. Although phagocytosis is a major functional ac tivity, it is only one of their several functions. The important role of phagocytes and other mononuclear cells in antibody forma tion, cell-mediated immunity, specific and non-specific resistance to microorganisms and tumor cells, as well as homeostatic adjust ments in general has become the focus of attention for many inves tigators.

This comprehensive treatise on the reticuloendothelial system is a project jointly shared by individual members of the Reticuloendothelial (RE) Society and bio medical scientists in general who are interested in the intricate system of cells and molecular moieties derived from those cells which constitute the RES. It may now be more fashionable in some quarters to consider these cells as part of what is called the mononuclear phagocytic system or the lymphoreticular sys tem. Nevertheless, because of historical developments and current interest in the subject by investigators from many diverse areas, it seems advantageous to present in one comprehensive treatise current information and knowledge con cerning basic aspects of the RES, such as morphology, biochemistry, phylogeny and ontogeny, physiology, and pharmacology as well as clinical areas including immunopathology, cancer, infectious diseases, allergy, and hypersensitivity. The circulatory system is usually considered to be composed of tubes of various diameters, characterized by collateral and terminal branches. There is also a tendency to treat blood vessels merely as conducting tubes in which the various structures of the wall act as mechanical pumps wlrich modify their diameter. This is, of course, not so. In fact, we know that blood vessels, and in particular arteries, are organs with personalities of their own and a particular susceptibility to several diseases. In addition, blood vessels differ in structure, according to their localization, and age at differing rates. The experimental work car ried out so far clearly confirms the data that have come from spontaneous human pathology; experimentally induced arterial lesions have a definite tendency to appear in certain arteries and not in others, depending on the experimental procedures used, and in each specific artery the lesions appear to have a specific location. We now know that the arterial wall is a metabo licallyactive structure, in which a number of enzyme activities have been clearly demonstrated. It possesses a sensitive vasa vasorum apparatus and a specific reactivity to various lesion-inducing stimuli. We must also remember that the arterial wall is in continuous contact with the blood circulating through the endothelial cells lining the vascular bed. It is obvious, therefore, that any variation in the circulating blood mass can modify the morphology as well as the function of the vessel wall.
