A new class of mutation was induced by X-radiation in Paramecium aurelia . These mutant segregants were called temperature-sensitive mutants ( ts mutants) since they can grow normally at 27° but are inviable at 36°. The maximum yield of ts mutant segregants after 2 kR of X-radiation was about 14% of the segregants viable at 27°. Crosses between persistent ts mutant segregants and wild-type stocks showed that the ts character is inheritable and recessive. Crosses between different ts mutant stocks revealed that these ts loci are widely spread over the genome of Paramecium. Certain subsidiary problems arising in the course of the work and the genetic applications of the ts mutations to genetic analysis of paramecia were discussed.
