Extracellular matrix (ECM) proteins are critical for the differentiation of normal mammary epithelial cells, but pronounced changes in the ECM occur during breast cancer. We have determined that estrogen receptor-positive breast cancer cells (MCF-7 and T47D) cultured on laminin have reduced, or lack, estrogen responsiveness. Estrogen-induced proliferation, following anti-estrogen treatment, did not occur on laminin (T4?D) or was significantly reduced (MCF-7), but did occur on collagen I or fibronectin. Laminin inhibited basal proliferation of MCF-7, T4?D and the ER- negative MDA MB23 1 cells, but did not inhibit IGF-I or EGF induced proliferation, indicating that tumor cells on laminin do not become refractory to mitogens.

Stromelysin-3 (ST-3), a member of the matrix metalloproteinase (MMP) family, is expressed in stromal cells surrounding invasive breast carcinoma cells. where it undergoes processing to its active form by an intracellular, proprotein convertase-dependent pathway. While strong correlations between disease progression and ST-3 expression suggest that the proteinase plays a critical role in breast cancer, human ST-3 is the only matrix metalloproteinase that does not express a marked ability to degrade components of the extracellular matrix (ECM). To further characterize the role of ST-3 in vivo, our attention focused on the analysis of lines of transgenic mice wherein the whey acidic protein (WAP) gene was used to direct human ST-3 expression to mammary tissue during late pregnancy through lactation. In these studies, high levels of the ST-3 transgene were generated in vivo which unexpectedly triggered a massive involution program. Surprisingly, ST-3 not only induced an apoptotic program, but also initiated changes in the underlying basement membrane in vivo. In addition, ST-3 expression also appeared to induce an exaggerated angiogenic response in involuting tissues. These data provide the first evidence that ST-3 can regulate mammary epithelial- ECM interactions and angiogenesis in vivo.

Angiogenesis is required for continued growth of most solid tumors, and provides a mechanism by which the stroma may continue to grow along with the tumor cells. Although endothelial cells are stromal cells, they express a number of epithelial characteristics including (i) epithelial-like tight junctions and junctional proteins; (ii) the ability to organize into flat­ tened tubular structures; (iii) the c-met receptor protein; and (iv) biologic responsiveness to SF. It is, perhaps, not surprising that vascular endothe­ lial cells may both produce and respond to SF in different situations. 'Epithelialness' may be defined in two ways: (i) expression of generic epithelial structures and proteins (eg., specialized junctions, junctional proteins [eg., cadherins, ZOl], cytokeratins); and (ii) production of specific differentiated products (eg., milk proteins by mammary epithelia, renin by renal tubular epithelia of the juxtaglomerular apparatus). Recent studies suggest that SF Ic-met signalling may mediate epithelia­ mesenchyme interconversion, in part by modifying some of the generic epithelial characteristics. Nusrat discusses the effects of SF on the epithelial junctional apparatus. Relatively little is known about whether and how SF regulates cell-specific differentiation.
