They are essential for vegetative propagation and survival of numerous species. The indeterminate growth of primary and lateral roots is supported of the quiescent centre and the stem cell niche around, with their functionality depending on several factors, e.g., auxin and cytokinin. In plant biology Arabidopsis thaliana is widely used as model plant because the availability of genetic information and culture protocols for studying root development. The investigation here described allowed to define the still unknown organization of the quiescent centre and stem cell niche in A. thaliana adventitious roots formed by intact hypocotyls and in vitro cultured stem thin cell layers. The aim was reached by the use of quiescent centre markers, auxin carriers, auxin biosynthesis genes, cytokinin localization and morphogenic and histological analyses of mutants.
