Book Description
Myofibroblasts (MFB) are found in most tissues of the body. They have the matrix-producing functions of fibroblasts and contractile properties that are known from smooth muscle cells. Fundamental work of the last decades has shed remarkable light on their origin, biological functions and role in disease. During hepatic injury, they fulfill manifold functions in connective tissue remodeling and wound healing, but overshooting activity of MFB on the other side induces fibrosis and cirrhosis. The present e-book "Liver myofibroblasts" contains 9 articles providing comprehensive information on "hot topics" of MFB. In our opening editorial we provide a short overview of the origin of MFB and their relevance in extracellular matrix formation which is the hallmark of hepatic fibrosis. Thereafter, leading experts in the field share their current perspectives on special topics of (i) MFB in development and disease, ii) their role in hepatic fibrogenesis, and (iii) promising therapies and targets that are suitable to interfere with hepatic fibrosis.

Book Description
Signaling Pathways in Liver Diseases, Third Edition again provides hepatologists and hepatology researchers with an expert overview of the complex and novel cellular/extracellular signaling pathways in the liver, and their role in liver diseases. The last few years have seen a great number of developments in this field, which in turn have led to new opportunities for innovative treatments; however, the intricacy of these pathways and their interactions continue to provide a real challenge for clinicians. This outstanding book compiles the emerging knowledge into a single expert resource, cataloguing and organizing it into an accessible and understandable format. With increased focus on the comprehension of cellular mechanisms involved in steatohepatitis, cirrhosis, and liver tumors, which has led to changes in the management of these diseases, this new edition also sees the introduction of exciting new chapters on key emerging areas such as: Autophagy Notch Pathway P13K/PTEN Signaling in Liver Diseases Sirtuins Hepcidin and Iron Epigenetic Regulation of Hepatic Stellate Cells and Liver Fibrosis Oxidative Stress and Signaling in the Liver. Professors Dufour and Clavien have assembled an all-star cast of chapter authors, each of whom has provided clear and appropriate illustrations to reinforce the text, with a key points box offering a concise and handy summary. Self-assessment questions and answers allow the reader to test their own knowledge. Signaling Pathways in Liver Disease, Third Edition is the perfect educational and reference tool to bridge the information exchange between the laboratory, the clinical ward, and the operating room, and an essential tool for the modern-day hepatologist.

Book Description
This book discusses clinical advances in hepatology, with a focus on metabolic diseases and chronic hepatitis C. The development of direct-acting antiviral (DAA) agents for the treatment of hepatitis C virus (HCV) infection in 2010 has completely transformed the management of this disease. This transformative nature of DAA therapy underpins the goal of the World Health Organization (WHO) to eliminate HCV infection as a public health threat by 2030. The advantages of using these therapies include high efficacy (sustained virological response rate >95%) with minimal side effects, good tolerability, easy drug administration (once-daily oral dosing) and short duration of treatment (8-12 weeks). The commercialization of second-generation DAA agents due to their high effectiveness, few side-effects and pangenotypic action. This transformative nature of DAA therapy underpins the goal of the WHO to eliminate HCV infection as a public health threat by 2030.

Book Description
Autosomal Dominant Polycystic Kidney Disease (ADPKD) is a highly prevalent hereditary renal disorder in which fluid-filled cysts are appeared in both kidneys. Main causative genes of ADPKD are PKD1 and PKD2, encoding for polycystin-1 (PC1) and polycystin-2 (PC2) respectively. Those proteins are localized on primary cilia and function as mechanosensor in response to the fluid flow, translating mechanistic stimuli into calcium signaling. With mutations either of PKD1 or PKD2, hyper-activated renal tubular epithelial cell proliferation is observed, followed by disrupted calcium homeostasis and aberrant intracellular cyclic AMP (cAMP) accumulation. Increased cell proliferation with fluid secretion leads to the development of thousands of epithelial-lined, fluid-filled cysts in kidneys. It is also accompanied by interstitial inflammation, fibrosis, and finally reaching end-stage renal disease (ESRD). In human ADPKD, the age at which renal failure typically occurs is later in life, however no specific targeted medications are available to cure ADPKD. Recently, potential therapeutic targets or surrogate diagnostic biomarkers for ADPKD are proposed with the advances in the understanding of ADPKD pathogenesis, and some of them were attempted for clinical trials. Herein, we will summarize genetic and epi-genetic molecular mechanisms in ADPKD progression, and overview the currently available biomarkers or potential therapeutic reagents suggested.
