The idea for this book arose during the 1985 Gordon Conference on "Mammary Gland Biology". New developments in the methodology of cell biology and the explosive growth of molecular biology had begun to impact upon our understanding of mammary gland growth and function. It seemed a propitious time for summarizing the current status of knowledge of the cell and molecular biology of mammary cancer and for attempting to outline future areas of concern and interest. The reviews presented here were completed by the Fall of 1986. Although new insights will surely continue to emerge, it is hoped that the material in this volume will form not only a current update but a basic core of information for future experiments. We have not attempted to cover all areas of mammary gland transformation. Those areas where recent detailed reviews are already available have been omitted. Also, the areas of normal gland development, cell ultrastructure, hormone responsiveness, chemotherapy and clinical aspects of mammary cancer have not been included. Instead, we have selected those areas where the development of new methodology, reagents and results have led to new ideas about mammary gland function and development as they are related to neoplasia.

1. 1 Scope of the Review This review was intended initially as a reference source for those interested in the origins and fITst descriptions ofthe defective avian sarcoma viruses. Quite a few of these viruses have been characterized in the past few years and their varied nomenclature according to source, discoverer, date of isolation or biological properties could result in some con fusion among those attempting to follow the literature. Information will be included on the molecular biology of the sarcoma viruses, rather more of which is available than when the review was fITst conceived, although in this respect the review will inevitably be out of date by the time of publication. If any bias of content is introduced, this will be towards a more detailed coverage of the author's own area of interest, the gene products of the defective sarcoma viruses. Rous sarcoma virus (RSV) serves as the model for much of this work and will frequently be referred to for comparative purposes, as will the mammalian defective transforming viruses. Recent reviews provide more complete coverage of these topics (l-4a). This review is complemented by a discussion ofthe avian acute leukaemia viruses, which appears elsewhere in this volume [5]. As will be seen, concentrating primarily on the defective sarcoma viruses and comparing them to RSV can be justified in terms of their biochemical properties as well as their similar biology.

Genetic analysis of microbial systems provided us with the foundation for un derstanding gene structure, expression, and regulation. It was long felt that the ability to generate mutants and conduct genetic studies in mammalian systems would prove to be equally useful. However, genetic analysis based on sexual systems is difficult in mammals because of the long generation times and the inability to perform controlled matings. As a result, genetic analysis of mam malian systems had to await the development of parasexual systems. This book is an attempt to bring together descriptions of a number of these parasexual systems. A common theme of all the parasexual systems is the transfer of genetic information from a defined source into a specific cell type. This volume deals with a number of methods of gene transfer into mammalian cells. The early methods of gene transfer involved transfer of relatively large amounts of genetic information. These include somatic cell hybridization, microcell fusion, and chromosome transfer, which constitute the first part of this book. Each of these methods has already proven to be of enormous value in arriving at a genetic understanding of the mammalian genome. Development of recombinant DNA methods, and the ability to introduce purified DNA into mammalian cells, has had a significant impact on our ability to dissect important aspects of mammalian gene expression and regulation. The second part of this book deals with gene transfer systems involving defined nucleic acid sequences.
