Looking to the future, the editors predict that the foundational information provided in this book -- combined with the advent of new tools and budding interests in use of non-rodent mammalian models -- will produce another major advance in knowledge regarding the biology of spermatogonia over the next decade. In particular, we anticipate that the core molecular machinery driving different spermatogonial states in most, if not all, mammals will be described fully, the extrinsic signals emanating from somatic support cell populations to influence spermatogonial functions will become fully known, and the capacity to derive long-term cultures of SSCs and transplant the population to regenerate spermatogenesis and fertility will become a reality for higher order mammals.

Spermatogenesis in the mammalian testis results in the daily production of millions of spermatozoa. This developmental process is founded upon the actions of a small population of spermatogonial stem cells (SSCs). As SSCs divide, their progeny must either remain an SSC to maintain the stem cell pool (self-renewal) or become an undifferentiated progenitor spermatogonium that proliferates prior to differentiating in response to retinoic acid (RA) and entering meiosis. Our laboratory is focused on understanding the mechanisms that regulate these fate decisions as well as defining the changes that occur downstream of RA that prepare spermatogonia for entry into meiosis. Currently, researchers have been unable to readily isolate pure populations of spermatogonia in the developing or adult testis. To address this, I have examined multiple transgenic mouse models to identify those with germ cell-specific expression of fluorescent reporter genes that will enable us to isolate spermatogonia using fluorescence-activated cell sorting (FACS). The results in this thesis describe these efforts, and document the identification of an excellent mouse model for future studies in our laboratory to understand the mechanisms underlying spermatogonial development. Spermiogenesis summarizes the process of sperm structure and development in a variety of organisms. It attempts to illustrate the structure of the organelles that characterize mature spermatozoa and to trace the morphogenesis of these organelles during sperm formation. This task is complicated by considerable variations in the structure of spermatozoa among different species and the lack of good morphological studies of sperm structure and spermiogenesis in many animal groups. The general morphological features of mammalian sperm structure differ in some ways from the features of insect sperm, the spermatozoa produced by insects appearing relatively more simple. When the functions and developmental regions of the organelles are considered, homologies between complex structures found in mammalian sperm and simpler insect sperm structures may be deduced even though the organelles in question are quite dissimilar in appearance. Such homologies will be stressed in an effort to present as general a picture of spermiogenesis as seems consistent with the species-specific variations that exist. This book will be a useful, stimulating reference for students of reproductive biology.
