The traditional method for evaluation of corn (Zea mays L.) germplasm for leaf-feeding resistance to European corn borer (ECB), Ostrinia nubilalis (Hübner), is conducted in the field by infesting plants with first instars and assessing the feeding damage three weeks after infestation. The main factor involved in leaf-feeding resistance to ECB in Midwest adapted cultivars has been linked to the presence of the chemical 2,4-dihydroxy-7-methoxy-1,4-benzoxaxin3-one (DIMBOA). This research involves developing a laboratory leaf extraction technique to find sources of ECB resistance that are not DIMBOA-based from eleven varieties of Peruvian corn. Corn leaves are collected from field-grown plants, then are freeze dried, milled, and stored in a freezer until needed for extraction.
