Des troubles de mémoire sémantique sont observés dans les pathologies neurodégénératives et plus particulièrement dans la maladie d’Alzheimer, dans la démence sémantique mais aussi dans la variante frontale de la démence fronto-temporale (vf-DFT). Toutefois, la nature des troubles et les mécanismes impliqués ne sont pas clairement établis dans ces différentes affections. L’objectif général de cette thèse est d’étudier la mémoire sémantique et de mieux comprendre ses déficits et leur évolution dans les trois pathologies. Un paradigme original reposant sur les effets d’amorçage sémantique pour différents types de liens sémantiques montre que dans la maladie d’Alzheimer et la démence sémantique, la dégradation des connaissances sémantiques génériques affecterait en premier lieu les attributs distinctifs des concepts. Ce profil de dégradation s’accorde tout à fait avec les modèles connexionnistes de la mémoire sémantique. Nous avons aussi étudié les processus cognitifs et les substrats neuronaux impliqués dans les performances aux épreuves de fluence verbale de patients souffrant de démence sémantique et de vf-DFT. Nos résultats confirment l’aspect multi-déterminé des épreuves de fluences et que les déficits sémantiques observés chez des patients vf -DFT sont le reflet de troubles exécutifs et de mémoire de travail. Enfin, nous montrons que dans la maladie d’Alzheimer et la démence sémantique, la dégradation sémantique affecte aussi les connaissances relatives aux personnes célèbres. Nos résultats indiquent que ce type de connaissance se dégraderait de la même façon que les connaissances génériques, c’est-à-dire en commençant par les caractéristiques les plus fines
