Multiphase flows are typically described assuming that the different phases are separated by a sharp interface, with appropriate boundary conditions. This approach breaks down whenever the lengthscale of the phenomenon that is being studied is comparable with the real interface thickness, as it happens, for example, in the coalescence and breakup of bubbles and drops, the wetting and dewetting of solid surfaces and, in general, im micro-devices. The diffuse interface model resolves these problems by assuming that all quantities can vary continuously, so that interfaces have a non-zero thickness, i.e. they are "diffuse". The contributions in this book review the theory and describe some relevant applications of the diffuse interface model for one-component, two-phase fluids and for liquid binary mixtures, to model multiphase flows in confined geometries.

This thesis presents work using multiphase, plug-based microfluidics with applications in fields related to energy and medicine. Reoccuring themes in the work are: (1) control of interfaces and transport properties in the plug-based microfluidic systems, (2) use of miniaturization and compartmentalization to enhance experimental throughput, (3) development of experimental designs and methods of analysis to maximize information from these systems, and (4) development and use of visual readout methods to enable fast and efficient data analysis. Functional fluorous surfactants that selectively bind histidine-tagged proteins were synthesized and utilized in plug-based microfluidics creating in effect, mobile self assembled monolayers, which enhanced protein crystallization. A genetic algorithm and microfluidic system were designed to discover and "evolve" homogeneous catalysts for partial methane oxidation using O 2. The system was enabled by selective gas and product transport and a visual detection system. A comprehensive method for analyzing digital PCR, and other digital assays, was developed. It used most probable number theory and incorporated multiple volumes for tunable control of dynamic range and resolution. The analysis and SlipChip platform were validated using PCR in both single and multivolume designs showing expected dynamic range and resolution. RT-PCR as well as isothermal methods using RPA and NASBA were also validated. This digital multivolume approach in SlipChip has potential for impact in developing viable quantitative point-of-care diagnostic tools for use in limited resource settings.
