Where do you begin to look for a recent, authoritative article on the diagnosis or management of particular malignancy? The few general oncology text books are generally out of date. Single papers in specialized journals are informative but seldom comprehensive; these are more often preliminary reports on a very limited number of patients. Certain general journals fre quently publish good in-depth reviews of cancer topics, and published sym posium lectures are often the best overviews available. Unfortunately, these reviews and supplements appear sporadically, and the reader can never be sure when a topic of special interest will be covered. Cancer Treatment and Research is a series of authoritative volumes which aim to meet this need. It is an attempt to establish a critical mass of oncology literature covering virtually all oncology topics, revised frequently to keep the coverage up to date, easily available on a single library shelf or by a single personal subscription. We have approached the problem in the following fashion. Translational control of rat mammary protein synthesis during pregnancy and lactation was investigated. A mammary cell-free system proved impractical due an inhibitor of translational initiation and a mammary- specific RNAse. However, using RNAse inhibitors, intact polyribosomes were prepared from mammary tissue. Both ribosome concentration and the proportion of palysomal ribosomes increased from late pregnancy to peak lactation. In contrast, the concentration of ribosomes and the proportion in polysomes both decreased when serum prolactin was experimentally depressed during lactation. Therefore, control of translation in vivo involves changes in both ribosome concentration and recruitment into polysomes. In mammary expiants, differences in protein synthesis per ribosome were measured after culture with insulin and cortisol, with or without prolactin. Translational responses to prolactin, were variable between different rats and different times of year. Responses to various hormone combinations were greater when expressed per unit DNA than per RNA suggesting that they involved both increased ribosome concentration and more efficient translation. Polysome analysis confirmed that the concentration of all ribosome species in expiants cultured with hormones exceeded that in expiants cultured without hormones but suggested no difference in the proportion of polysomal ribosomes. Therefore, alterations in translational efficiency in vitro were due to concomitant changes in translational initiation, elongation and termination. Insulin-like growth factor-1 and epidermal growth factor (IGF-1 and EGF) may modulate mammary proliferation and differentiation. In the presence of cortisol and prolactin, the response of translational efficiency to IGF-1 was, like that to insulin, dose dependent over a wide concentration range with a probable minimum effective concentration of lOng/mL. This suggested that each hormone acted through its own receptor. In contrast, EGF stimulated translational efficiency independently of accompanying hormones or concentration between 1 and 20ng/mL Results indicate that both growth factors affect translational efficiency in expiants.
