The unicellular eukaryote Saccharomyces cerevisiae can be used as a model system to study ubiquitin-dependent uptake of a signaling receptor. A screen for mutants defective in receptor uptake identified a number of possible candidates genes in this pathway. One of the isolated mutants was characterized further, revealing a novel role for a conserved kinase cascade in regulating endocytosis.

The finding that monoubiquitin serves as an autonomous internalization and the identification of ubiquitin-binding proteins that function in endocytosis validates the importance of the role of ubiquitin in endocytosis. One hypothesis how ubiquitin signals internalization is that specific endocytic proteins recognize and recruit the ubiquitinated cargo to the endocytic pathway. Because the ubiquitin binding domains of epsins and Vps27 are required for ubiquitinated cargo sorting at the internalization and multivesicular sorting step, they may be the potential adaptor proteins that recognize ubiquitinated cargo at distinct cellular compartments.
