Tomato (Lycopersicon esculentum Mill.) is the second most consumed vegetable aft er potato in the world and one of the most important vegetables grown in Lebanon under both greenhouse and field conditions. Tomato is widely grown for its tast e, flavor, color, and health benefits. Since 1976, Tomato yellow leaf curl virus (TYLCV), has been causing economic damage to tomato in Lebanon, and has become the major limiting factor for tomato production in the coastal areas especially in the fall growing season, where yield loss may reach 100% if seedlings are inf ected early in the growing season. Lebanese tomato Landraces are highly suscepti ble to TYLCV and the development of resistant cultivars is considered as an attr active option for management of TYLCV infections. Tomato yellow leaf curl diseas e may be induced by several species or strains of TYLCV that are all transmitted by the whitefly vector Bemisia tabaci. A survey was undertaken, during summer 2 004 and spring 2005, in the main tomato growing areas. Results indicated that TY LCV in Lebanon was caused by either single or mixed infections with the mild str ain (TYLCV-Mld) and/or the Israeli strain (TYLCV-Is), while the Almeria strain w as not detected. Tomato populations resulting from crosses of four Lebanese toma to landraces with two wild Lycopersicon species (L. chilense and L. hirsutum) we re evaluated for their resistance against TYLCV. Three field trials were conduct ed in plastic tunnels, in Damour South of Beirut. Prior to transplanting, all se edlings were artificially inoculated with TYLCV-Is and TYLCV-Mld using virulifer ous whiteflies. Eight tomato plants belonging to the F2 population were screened for resistance to TYLCV during the first field trial. Plants showing mild or no symptoms and the desirable horticultural characteristics were selected, their s eeds collected and used to evaluate further progenies. Virus titer (concentratio n) in the majority of tolerant/resistant tomato genotypes was lower than the cor responding susceptible genotypes as determined by nucleic acid hybridization usi ng chemiluminescent detection system. Tomato (Lycopersicon esculentum Mill.) is one of the most important vegetables grown in Lebanon. In 1976, it was reported that tomato yellow leaf curl geminivirus (TYLCV) causes economic damage on tomato in Lebanon especially in the Fall growing season, whereby incidence varied from 28-50% in September to 85-90% in December at most locations. This led Lebanese farmers to reduce or stop growing tomatoes during the Fall season. During the last few years, several private firms and public research institutions initiated breeding programs in order to develop tolerant or resistant tomato varieties. The following wild tomatoes were used as sources of resistance: Lycopersicon pimpinellifolium, L. chilense, L. peruvianum, L. hirsutum and L. cheesmanii. This study focused mainly on two aspects: a- screening tomato cvs. for resistance to TYLCV by including some commercially released hybrids, and participating in an EEC-funded research network for breeding tomato lines resistant to the virus; b- diagnosis and detection of TYLCV using DNA hybridization techniques, and the application of non-radioactive DNA probes for this purpose. Ten tomato varieties were screened for resistance to TYLCV at the American University of Beirut (AUB). Four determinate varieties were found to be highly tolerant to the virus, producing milder symptoms and relatively high yield as compared to the susceptible controls most commonly used in Lebanon. Only one indeterminate tolerant variety was tested, and proved to exhibit milder disease symptoms but the yield was reduced significantly. In the context of a breeding program aiming at incorporating new genes of resistance to TYLCV in cultivated tomato species, the composite segregating population CHEPERTYLC 92 was screened for resistance. The CHEPERTYLC 92 program aims at pooling the resistances issued from the two populations: CHETYLC 89 and PERTYLCEG 90, which originate respectively from the two wild lines of tomato: L. cheesmanii LA 1401 and L. peruvianum. Eleven tomato plants were selected out of 363 plants of the segregating population CHEPERTYLC 92, and seeds were collected to be included in future breeding programs. Molecular hybridization techniques for routine testing of plant viruses have gained worldwide acceptance in recent years, and were found to be more sensitive than ELISA. Therefore, four different extraction techniques of TYLCV were tried, in addition to tissue blots, and were all tested under three different hybridization techniques. The two extraction methods using 10 SSC-0.01% SDS, pH=7 and 0.4 N NaOH respectively were found to be effective, practical, fast and sensitive. Excellent results were obtained using the tissue blot technique, which consists of simply blotting the cross section of an infected tissue (stem or leaf) on a membrane and then subject it to hybridization. High concentrations of virus particles were detected in tissue blots of infected stem sections as compared to infected leaf sections. Two of the hybridization techniques tested were successful in detecting as little as 0.45 pg of viral DNA; the two hybridization buffers used were respectively: 50% formamide- 5 SSC, pH=7 and 50% formamide- 0.125M Na2HP04, pH=8. However, the first buffer gave slightly better results than the second.
