The National Institute of Dental Research sponsored a workshop on "Genetically Engineered Vaccines: Prospects for Oral Disease Prevention," held at the National Institutes of Health (NIH) on November 6-8, 1991. The purpose of the workshop was to convene molecular biologists and immunologists to address the state of the science in vaccine development and to explore the potential of developing vaccines for prevention of oral diseases. The goal was to elicit new research initiatives and recommendations for vaccine development with emphasis on the prevention of oral diseases and diseases affecting the orofacial tissues. The workshop was attended by more than 100 persons who heard 30 presentations, and the speakers provided the papers for this volume. The workshop focused on the following topics: oral diseases and host immune responses, update on vaccines and vaccine development, vaccines and the mucosal immune system, optimizing mucosal and systemic immune responses, delivery systems and immune analysis, target antigen selection and vaccine development, immunological correlates of protection and future direc tions/recommendations. Three key areas were identified: Optimizing the Mucosal Immune Response, Antigen Delivery Systems, and Target Antigens and Immunological Correlates of Protection. The summary and recommendations from these deliberations is included at the end of this volume.

Infectious diseases are still a major threat to public health. The vaccine remains the most important and safer way of combating infectious diseases. Therefore, there is an urgent need to look for new ways of vaccine generation that can cut down production costs and processing time. The development of rDNA technology allows the expression and purification of proteins in unlimited quantity, thus opening an avenue for the development of protein-based vaccines as a prophylactic measure against infectious diseases. Recombinant therapeutic proteins derived from biological sources, including mammalian cells, microorganisms, suspension cultures, or genetically modified organisms by employing biotechnological processes, are widely used in clinical applications, especially for the treatment and prevention of human or veterinary infections. Since the development of human insulin by utilizing recombinant DNA technology in E. coli in the 1980s, the recombinant therapeutic protein production field has significantly grown and gained major attention. Plant molecular farming, the production of biologics in plant cells or transgenic plants, could offer a cost-effective adaptable strategy to produce biologics, particularly in low-resource settings. Molecular farming-based strategies could provide an alternative strategy to traditional biologics or vaccine production, enabling rapid development, effective deployment, and safe administration of vaccines. The concept of PMF was initially documented three decades back when recombinant growth hormone was produced in tobacco and sunflower plants. Even with such a long history of significant advantages and several proof-of-concept studies, very few plant-derived therapeutic products have been clinically translated. However, the recent progress around the approval of the plant-based biopharmaceutical ZMapp by the FDA, Medicago’s COVID-19 vaccine by Health Canada, and the safety of a few plant-derived vaccines in clinical trials have displayed the potential of a plant platform for biopharmaceutical production. Furthermore, constant efforts are being made with respect to the optimization of expression techniques, downstream processing, and defining standard guidelines for plant products to make an impact of plant-derived products on an array of applications.

Abstract: Abstract of thesis entitled PRODUCTION OF TRANSGENIC PLANT-DERIVED VACCINES VIA PLASTID TRANSFORMATION TECHNOLOGY Submitted by Lee Yuk Ting for the degree of Master of Philosophy at The University of Hong Kong in July 2004 With the advent of genetic engineering in higher plants, the need for very large quantities of therapeutic protein at low cost, and the desire to have heat-stable edible vaccines directed at human and animal diseases, transgenic plant-derived vaccines offer a new strategy for the development of safe, inexpensive vaccines against infectious diseases. The first success of plastid transformation in tobacco in 1990 has opened up the opportunities for genetically modifying plastids in higher plants for high level expression of biopharmaceuticals, such as antibodies and vaccines for oral administration. Since each plant cell contains up to 10,000 copies of identical plastid genome, plastid engineering should result in very high levels of transgene expression. In addition, due to the prokaryotic feature of the plastids, engineering the plastid genome bears the advantages of expressing multiple genes as operons without the problem of positional and epigenetic effects affecting transgene activity. Therefore, plastid transformation technology is a promising alternative to the conventional nuclear transformation. The objectives of this research are: 1) to examine if plastids can be used to produce edible vaccines suitable for uses against Epstein-Barr (EBV) and Hepatitis B Virus (HBV), and 2) if such a production can be regulated through a chemically inducible gene expression system. The genes encoding the early antigen of the Epstein-Barr virus and the Hepatitis B surface antigen (HBsAg) are introduced into tobacco plastids via biolistic bombardment techniques. Simultaneously, an alcohol-inducible alc gene expression system is established and integrated into the tobacco nuclear genome for controlling plastid expression of antigen genes. It was found that chloroplast- derived antigen against EBV could be successfully produced in transplastomic tobacco using a constitutive plastid-derived promoter. However, constitutive expression of Hepatitis B surface antigen (HBsAg) apparently results in growth retardation and lethality of the plants. By adapting the ethanol inducible (alc) gene expression system to control engineered plastid operons, it is possible to regulate a marker gene (uidA) expression and the production of antigen (HBsAg) in plastids upon ethanol administration. The technology developed here could contribute to a much more cost-effective system for vaccine production system in the future.
