Sequence-independent DNA binding and the mechanism of translocation in B-family polymerases were examined in DNA substrate complexes of phi29 DNA polymerase. These complexes included the translocated binary complex and the post-insertion pre-translocation ternary complex. Comparison of these structures revealed a mechanism of translocation, different from the mechanism for A-family polymerases, based on the coordinated movement of two conserved tyrosine residues.
