Scientific Frontiers in Developmental Toxicology and Risk Assessment reviews advances made during the last 10-15 years in fields such as developmental biology, molecular biology, and genetics. It describes a novel approach for how these advances might be used in combination with existing methodologies to further the understanding of mechanisms of developmental toxicity, to improve the assessment of chemicals for their ability to cause developmental toxicity, and to improve risk assessment for developmental defects. For example, based on the recent advances, even the smallest, simplest laboratory animals such as the fruit fly, roundworm, and zebrafish might be able to serve as developmental toxicological models for human biological systems. Use of such organisms might allow for rapid and inexpensive testing of large numbers of chemicals for their potential to cause developmental toxicity; presently, there are little or no developmental toxicity data available for the majority of natural and manufactured chemicals in use. This new approach to developmental toxicology and risk assessment will require simultaneous research on several fronts by experts from multiple scientific disciplines, including developmental toxicologists, developmental biologists, geneticists, epidemiologists, and biostatisticians.

At the request of the National Institute of Environmental Health Sciences, the National Research Council assembled a committee to identify the benefits of toxicogenomics, the challenges to achieving them, and potential approaches to overcoming such challenges. Genotoxic compounds in the diet represent a truly international problem. This problem was recognized in Japan and the United States of America only a few years ago by the application of in vitro mutagenicity tests to prepared and unprepared components of our diet. Now new discoveries are appearing every day concerning genotoxic compounds in our food supply from all over the world. It seems likely that naturally occurring genotoxic compounds in the diet as well as genotoxic compounds formed during storage, preparation, and digestion of the food are important for the induction of cancer in humans and of hereditary changes in human germ cells. Other scientific data indicate that dietary constituents have the potential to modify the genotoxic effect of these compounds in humans. These findings indicate new possibilities for the prevention of human diseases of genotoxic origin through relevant changes in the composition of the diet. With this background, and due to the rapid accumulation of new data in the field, it was approved by the Board of the International Association of Environmental Mutagen Societies that one of the satellite meetings of the Fourth International Conference on Environmental Mutagens in Scandinavia 1985 should cover the aspects of mutagens and carcinogens in the diet. The satellite symposium on "Genetic Toxicology of the Diet" took place in Copenhagen, June 19-22, 1985. The present volume contains the proceedings of this symposium.
