NEUROTOX '91 was the fourth meeting in a series which started in 1979. The '91 meeting, like its predecessors, was held under the patronage of the Society of Chemical Industry, and despite the unfortunate proximity of hostilities in the Arabian Gulf attracted a truly international mix of industrial and academic pesticide scientists. This volume contains the text of invited papers read at the meeting and presents the dramatic developments which so excited those who attended. The potential of molecular neurobiology for gaining knowledge of target sites for neurotoxicants is now starting to be realised. These studies, in conjunction with developments in molecular imaging and modelling, provide new oppor tunities for chemists and biologists to gain insights into molecular interactions underlying intoxication. Molecular techniques have also enabled rapid ad vances on a second front, where the cloning of genes controlling pesticide resistance should have a profound impact on our understanding of this commercially important problem. The understanding of molecular events will undoubtedly be vital in future developments in chemical control of pests; however, the value of understand ing the way in which the nervous system controls behaviour and how behaviour can be modified by chemicals of bath synthetic and natural origins was highlighted. Natural products and their synthetic analogues have continued to provide new and interesting molecules which are already proving their worth as tools for the neuroscientist and may offer leads for commercial synthesis.

The development of pesticide resistance in arthropod pests, plant pathogens and weeds can be viewed and studied from two contrasting perspectives. At a fundamental level, resistance provides an almost ideal example of adaptation to withstand severe environmental stress. Population geneticists, biochemists and, most recently, molecular biologists have cast considerable light on the nature of this adaptation in diverse taxonomic groups, and on factors determining its selection and spread within and between populations. Unlike most evolutionary phenomena, however, resistance is also of immediate practical and economic significance. Not only has the number of resistant species continued to increase inexorably, but there has been an alarming increase in the severity and extent of some resistance problems. Cases of organisms resisting virtually all available pesticides are by no means uncommon, and pose a formidable challenge in view of present difficulties in discovering and developing novel chemicals. Although most occurrences of resistance were initially monofactorial, resistance now frequently involves a suite of coexisting mechanisms that protect organisms against the same or different pesticide groups, and may even predispose them to resist new, as yet unused chemicals.

Neurotoxicology is a broad and burgeoning field of research. Its growth in recent years can be related, in part, to increased interest in and concern with the fact that a growing number of anthropogenic agents with neurotoxic potential, including pesticides, lead, mercury, and the polytypic bypro ducts of combustion and industrial production, continue to be spewed into and accumulate in the environment. In addition, there is great interest in natural products, including toxins, as sources of therapeutic agents. Indeed, it is well known that many natural toxins of broadly differing structure, produced or accumulated for predatory or defensive purposes, and toxic agents, accumulated incidentally by numerous species, function to perturb nervous tissue. Components of some of these toxins have been shown to be useful therapeutic agents and/or research reagents. Unfor tunately, the environmental accumulation of some neurotoxic ants of anthropogenic ori gin, especially pesticides and metals, has resulted in incidents of human poisoning, some of epidemic proportion, and high levels of morbidity and mortality. Furthermore, an increasing incidence of neurobehavioral disorders, some with baffling symptoms, is confronting clinicians. It is not clear whether this is merely the result of increased vigi lance and/or improved diagnostics or a consequence of improved health care. In any case, the role of exposure to environmental and occupational neurotoxic ants in the etiology of these phenomena, as well as neurodegenerative diseases, is coming under increasing scrutiny and investigation.

This book contains the proceedings of the "First International Symposium on Insect Pheromones," which was held at Wageningen, The Netherlands, from March 6 to March 11, 1994. Eighty participants from 17 countries attended the symposium, which turned out to be a unique forum for the exchange of the latest worldwide findings on insect pheromones, an opportunity to discuss and debate unsettled issues, and a mechanism to define new directions in pheromone research and foster interdisciplinary collaborations. The meeting comprised five sessions representing the breadth of disciplinary interest in pheromones, a typical charac teristic of this research area. In the sessions the following topics were presented: (1) control of pheromone production (organized by W. L. Roelofs), (2) sensory processing of pheromone signals (T. L. Payne), (3) neuroethology of pheromone mediated responses (T. C. Baker), (4) use of pheromones in direct control (A. K. Minks and R. T. Card6), and (5) evolution of pheromone communication (c. LOfstedt). All sessions started with a series of 30-minute lectures, after which ample time was reserved for discussion. In each session some participants were asked to serve as discussants and to initiate and stimulate discussion, and a rapporteur was recruited to make notes of these discussions and to summarize the general trends emerging from the session. The specific topic, “Isoquinolines And Beta-Carbolines As Neurotoxins And Neuroprotectants – New Perspectives In Parkinson`S Disease Therapy,” was chosen in light of accumulating neurobiological evidence indicating that, in addition to exogenous neurotoxins (e.g., 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine [MPTP]), endogenous compounds may play an important role in the most common neurodegenerative disorders (e.g., Parkinson`s disease). Two groups of amine related compounds, which appeared chemically like MPTP, were detected in human brain and cerebrospinal fluid (CSF): ß-carbolines (BCs) and tetrahydroisoquinolines (TIQs). These are heterocyclic compounds formed endogenously from phenylalanine/tyrosine (TIQs) and tryptophan, tryptamine, and 5-hydroxytryptamine (BCs), respectively and exert a wide spectrum of psychopharmacological and behavioral effects. The TIQs and BCs may bind to their own high affinity sites on neuronal membranes associated with or located close to the receptors of neurotransmitters. Research on TIQs and BCs is stimulated also by their possible role in pathological conditions, especially parkinsonism and alcoholism. Recently, clinical interest has been spurred by their role as neuroprotective, and even neurorestorative, anticonvulsant and antiaddictive, substances. In this book we are going to summarize, for the first time, the results from behavioral, neurochemical and molecular experiments, which demonstrate a wide spectrum of TIQs and BCs effects - from their rather mild neurotoxic actions to the important neuroprotective and antiaddictive properties. Additionally, the recent results of experimental studies in vivo have allowed a much better understanding and simultaneous comparison of the neurochemical and molecular mechanisms underlying the neuroprotective and neurotoxic actions of endogenous TIQs and BCs and have pointed to the possibility of their therapeutic applications in neurodegenerative diseases such as Parkinson's disease.
