A study was undertaken to isolate and characterize avian metapneumovirus (aMPV) causing swollen head syndrome in chickens. Out of 21 oropharyngeal swab samples collected, only one sample from eight weeks old commercial layers was positive for aMPV N gene specific RT-PCR. The positive sample was inoculated in vero cells and isolated after four blind passages. The virus was identified by Agar gel precipitation test with reference aMPV hyperimmune serum. Further it was confirmed by N gene specific RT-PCR. Molecular subtyping based on G gene specific RT PCR revealed that the isolate belong to aMPV subtype A. The isolate was shown to have a homology of 96.4 per cent with the European isolate (#8544) and 89.7 to 100 per cent with other isolates of Europe, South Korea and Brazil (SHS/BR/669/03). Seroprevalence study of 1745 serum samples collected from 69 flocks using ELISA revealed that 925 samples from 48 flocks (69.56 %) were positive for aMPV antibodies with a GMT of 3513 114. The inactivated mineral oil adjuvant vaccine prepared from the isolate induced effective seroconversion after booster vaccination."

Numerous field isolates of avian infectious bronchitis virus (IBV) were recovered from flocks during a limited avian influenza outbreak on the Delmarva Peninsula during the winter of 2004. A number of these isolates came from flocks with elevated mortality rates. Ten isolates recovered from farms with mortality of 4 per 1000 per day to 7.5 per 1000 per day were characterized. Tracheal swab samples from flocks being tested for the avian influenza virus were sent to the Allen Laboratory of the University of Delaware for virus isolation attempts performed in specific pathogen free (SPF) embryonated eggs. Many of the inoculated embryos showed stunting and curling; lesions indicative of possible infection with IBV. The presence of IBV in suspected isolates was verified by reverse transcription-polymerase chain reaction (RT-PCR). Identification of the IBV genotype was determined. A fragment of the S1 subunit of the spike glycoprotein of isolates positive for IBV was amplified and sequenced. An alignment was performed between these S1 gene sequences and sequences of reference strains of IBV (Arkansas, Connecticut, Massachusetts, and Delaware 072) and commercial vaccines for IBV used along the peninsula. A total of 101 isolates were successfully sequenced. Eighty two of the isolates were of the Arkansas genotype (92.6% to 100% amino acid similarity). Four of the isolates were found to be the Connecticut genotype (97.1% to 98.9% amino acid similarity). Fourteen of the isolates had an amino acid similarity closest to the Massachusetts genotype (94.6% to 100% amino acid similarity). The remaining IBV isolate was identified as Delaware 072 (90% amino acid similarity). Additional characterization was conducted on a group of ten isolates. All of the isolates came from flocks experiencing mortality of at least 4 birds per 1000 per day. Non-vaccinated broiler chicks were inoculated with the ten isolates under study in order to determine pathogenicity. A vaccination-challenge of immunity study of specific pathogen free (SPF) chickens was performed to determine the effectiveness of vaccination against challenge of isolates of the same genotype. The presence of chicken anemia virus (CAV) in isolates was determined by a nested PCR.

Avian infectious bronchitis virus (IBV) is the causative agent of the disease infectious bronchitis in chickens which occurs globally and causes significant economic damage. The control of IBV remains a challenge and vaccination against IBV is performed extensively. Because of the restricted species tropism of IBV, vaccine strains have to be cultivated in embryonated chicken eggs (ECEs), which represent a laborious and expensive propagation method. The possibility of in vitro propagation of IBV vaccines in cell culture is highly desired. Usually, IBV strains cannot be propagated in cell lines apart from one exception: The Beaudette strain allows in vitro propagation in cell lines and thus has already been extensively characterized. Certain amino acids in the Beaudette spike protein were identified to be responsible for the ability of Beaudette to infect cell lines. However, so far cell culture-adapted IBV strains based on the Beaudette strain have failed to confer sufficient protection against challenge because of the apathogenic nature of the Beaudette strain. IB66HP, an IBV strain that was adapted to EB66® cells, represents a novel IBV strain with cell culture tropism. The objective of this thesis was to determine the molecular determinants responsible for the cell culture tropism of IB66HP in comparison to the determinants described for IBV Beaudette. An IBV reverse genetic system was used to generate several recombinant IBVs (rIBVs) for in depths analysis of the influence of the IB66HP spike protein sequence in the context of the egg-restricted IBV H52.
