The symposium reviewed progress in the understanding of scientific and biomedical problems from a biochemical, biophysical, pharmacological, and toxicological perspective. Volume I contained 117 contributions made by researchers at the symposium, which were organized into three sections (Sections I-III). This second volume contains 122 contributions, divided into four sections (Sections IV-VII). The papers on Section IV examine the metabolic fate of oxygenated compounds. Section V provides studies on microsomal enzymes and lipid metabolism. Section VI includes papers on microsomal enzymes and toxicity of foreign compounds. This book is one of a GANN Monograph on Cancer Research series and is based on a Japanese Cancer Association-sponsored symposium entitled "Multiplicity of Chemical Carcinogen-Activating Enzymes· with Particular Emphasis on Cytochrome P-450" which was held in Tokyo on November 21, 1983. The principal role of microsomal cytochrome P-450 in the metabolic activation of various chemical carcinogens in animal cells has been well established. The activated chemical carcinogens induce mutations of the cells, which is an essential part of the initiation of chemical carcinogenesis. Since microsomal cytochrome P-450 in various animal tissues includes multiple molecular species with different substrate specificities and different responses to chemical inducers, knowledge of the molecular and catalytic properties and the inducibilities of those cytochrome P-450 forms is surely a most important base from which to elucidate the mechanism of chemical carcinogenesis. The focus of the symposium was this area of molecular diversity and inducibility of microsomal cytochrome P-450 in animal tissues. The mechanism of activation of various types of chemical carcinogens by cytochrome P-450 and related enzymes was another main topic. The editors, who were also the symposium organizers, hope these summaries of current research in this field in Japan will stimulate future development of studies on the mechanism of chemical carcinogenesis.

The concept that detoxication is the inevitable result of biotransformation of xenobiotic compounds by mammalian systems has undergone modification since it was first described. Indeed, despite the fact that R. T. Williams popularized the notion, he was among the first to caution that it was not possible to predict the biological activities of the resulting metabolites. It has become increasingly apparent in recent years that not only do many metabolites of drugs and other chemicals display biological activity but also in many instances these metabolites play an important role in initiating several forms of cancer and are the cause of a variety of types of toxicity. Thus it seems appropriate to collect in one volume a series of reports outlining advances in the study of the formation of chemically active intermediary metabolic products of chemicals, mechanisms of toxicity or carcinogenesis, and pathways for true detoxication of these chemicals. The work of R. T. Williams, beginning in the late 1920s and early 1930s, marked the first concerted effort to understand the biotransformation of foreign chemicals in animals. He investigated the metabolic pathway of numerous compounds in a wide variety of animal species while training large numbers of our colleagues who have been responsible for further advances in biochemistry, pharmacology, and toxicology. The aim of the editors of this volume is to use basic and ap plied studies in the field of mutagenesis to approach a problem of especial concern. The problem is that of the usage of toxic chemi cals, particularly agricultural chemicals, in ever-increasing quan tities in those parts of the world that feed the most people. Agri cultural chemicals that are in use in Pakistan are emphasized here. These are the same chemicals that are in use throughout the develop ing world, although the quantities of the different types that are used may vary from country to country, and from region to region within countries. A number of these chemicals can no longer be sold in Europe or in the United States, and it is often difficult to iden tify a scientific reason as to why they are sold at all. It is ironic that toxic chemicals are used as a humanitarian device to rid the world of Pestilence and Famine -- two of the Horsemen of the Apocalypse. If we do not wish poisonous chemicals to become the fifth Horseman of the Apocalypse, then we must begin now to identify and regulate the large-scale usage of toxic sub stances everywhere.
