Book Description
Site Specific and Global Epigenomic Editing: Principles and Applications, Volume 33 highlights the expanding role that epigenetics plays in pathogenesis, the full range of sites for possible epigenetic modification, and the potential to target these sites to modulate disease progression. The book answers key questions, including which of the hundreds of epigenomic marks and features are relevant, and which are likely to serve a role in regulating gene expression, cell behavior and phenotypic outcomes. This volume is the first to provide a thorough, conceptual introduction to epigenetic regulation and a thorough accounting of sites for possible epigenetic edits and disease modulation. Included protocols and applied case studies examine early disease targets and likely pathways of therapeutic significance while also outlining common challenges in the field. The book addresses its topic in a comprehensive, but accessible manner, highlighting practical tools and burning questions remaining in the field to enable fresh research. - Offers a full, conceptual and applied framework for implementing epigenomic editing in biological research and drug discovery - Discusses fundamental theory, experimental design and applied approaches alongside common research challenges and bioethical concerns - Examines epigenomic editing's potential role in the modulation of developmental, neuropsychiatric and immunological disorders, as well as in viral infections and cancer

The DNA, RNA, and Histone Methylomes
Author: Stefan Jurga
Publisher: Springer Nature
ISBN: 3030147924
Category : Medical
Languages : en
Pages : 624
Book Description
This book reviews the chemical, regulatory, and physiological mechanisms of protein arginine and lysine methyltransferases, as well as nucleic acid methylations and methylating enzymes. Protein and nucleic acid methylation play key and diverse roles in cellular signalling and regulating macromolecular cell functions. Protein arginine and lysine methyltransferases are the predominant enzymes that catalyse S-adenosylmethionine (SAM)-dependent methylation of protein substrates. These enzymes catalyse a nucleophilic substitution of a methyl group to an arginine or lysine side chain nitrogen (N) atom. Cells also have additional protein methyltransferases, which target other amino acids in peptidyl side chains or N-termini and C-termini, such as glutamate, glutamine, and histidine. All these protein methyltransferases use a similar mechanism. In contrast, nucleic acids (DNA and RNA) are substrates for methylating enzymes, which employ various chemical mechanisms to methylate nucleosides at nitrogen (N), oxygen (O), and carbon (C) atoms. This book illustrates how, thanks to there ability to expand their repertoire of functions to the modified substrates, protein and nucleic acid methylation processes play a key role in cells.
