Experts explore the influence of trace metals on the pathogenesis of infectious diseases. Many parts of the world in which common infectious diseases are endemic also have the highest prevalence of trace metal deficiencies or rising rates of trace metal pollution. Infectious diseases can increase human susceptibility to adverse effects of metal exposure (at suboptimal or toxic levels), and metal excess or deficiency can increase the incidence or severity of infectious diseases. The co-clustering of major infectious diseases with trace metal deficiency or toxicity has created a complex web of interactions with serious but poorly understood health repercussions, yet has been largely overlooked in animal and human studies. This book focuses on the distribution, trafficking, fate, and effects of trace metals in biological systems. Its goal is to enhance our understanding of the relationships between homeostatic mechanisms of trace metals and the pathogenesis of infectious diseases. Drawing on expertise from a range of fields, the book offers a comprehensive review of current knowledge on vertebrate metal-withholding mechanisms and the strategies employed by different microbes to avoid starvation (or poisoning). This book originated in a series of cross-disciplinary conversations in the years 1984-1990 between the editor, who is a physician-researcher involved in clinical and laboratory research, and a dioxin toxicologist. During the years in which the conversations took place, an extraordinary amount of new scientific literature was published related to dioxins, defined for purposes of this text as the chlorinated dibenzo-p-dioxins, dibenzofurans, polychlorinated biphe nyls (PCB's) and other compounds that are structurally and toxicologically similar to 2,3,7,8-tetrachlorodibenzo-p-dioxin (2,3,7 ,8-TCDD), the most extensively studied and most toxic of this group of chemicals. Dioxins also began to interest not only chemists and toxicologists, but also specialists from diverse disciplines such as wildlife and environmental science, immunology, neuroscience,public health, epidemiology, med icine, government, law, sociology, and journalism. Specialists from such varied disciplines, while familiar with their own literature, frequently did not have time to follow the dioxin literature outside their specialty area. In addition, each specialty had unique knowledge, methods, and perspectives. Cross disciplinary conversation was necessary, but all too frequently, specialists from the various disciplines did not speak the same language, resulting in misunderstanding.

Homeostasis. The health of an organism is influenced by external and internal changes that may lead to the loss of homeostasis. Under healthy conditions organisms compensate these changes. If compensation fails disease ensues. Attention will be paid to lifestyle, environmental changes, genetic makeup and health system. It will be answered how lifestyle, environment, genetic makeup and social conditions help to maintain or upset the biological balance and lead to cancer. Tumor formation. To understand this process the transfer of intracellular and the pathways of extracellular information (signal transduction) will be reviewed briefly. Loss of cellular balance may lead to cell death (.e.g. apoptosis) or to rapid cell growth of cells leading to tumor formation. Metastasis. Animal tumor models serve to understand the spread of the primary tumor cells to distant locations of the organism. Different types of tumors and metastases will be reviewed.
