Part 1: Calibration models are important for the use of Spanner-type psychrometers, but they have not been adequately evaluated. A non-zero intercept in the regression between psychrometric electrical output (EO) and water potential (WP) is commonly observed, but incompatible with the temperature correction factor (TCF) model, most often used to account for the temperature-dependence of the EO/WP relation. Calibration errors (mean absolute [predicted-observed] WP) for 10 commercial, in-situ, temperature-compensated psychrometers, were compared over a temperature and WP range of 10-40°C and -2.5 to 0 MPa, respectively, using the current commercial TCF model, a modified TCF model, two empirical models, and a mechanistic model. For TCF models, a psychrometer-specific, linear, EO/WP relation was determined at 25°C, but models differed in whether extrapolation to other temperatures was based only on the 25°C data or on data obtained at all temperatures, and whether model parameters were determined for each psychrometer (individual-fit) or all psychrometers (group-fit). Measurement error was estimated at 0.042 MPa, and only the individual-fit modified TCF model exhibited an error (0.048 MPa) which was not statistically different from the measurement error. Individual-fit errors for other models ranged from 0.058 to 0.100 MPa. Group-fit errors ranged from 0.067 (current commercial TCF) to 0.109 MPa, with very little difference among models, indicating the current commercial model as adequate for a group-fit approach. Part 2: The hydraulic connection between the grape (Vitis vinifera ‘Chardonnay’) berry and the parent vine generally decreases over berry development between pre- and post-veraison, and it is important for understanding the response of berry size and expansive growth to changes in vine water status. However, the reported pathway hydraulic conductance (estimated using excised samples) varied greatly and spanned over one order of magnitude. In addition, it is unclear where the decrease of hydraulic connection occurs along the water transport pathway between the berry and the parent vine. This study aims to: 1) estimate the pathway hydraulic conductance in situ, and 2) locate the site responsible for the decrease of hydraulic connection in situ. We manipulated the stem water potential (SWP) of a grapevine by drying, root pressurization/depressurization, and re-watering, and measured the responses of berry diameter and the turgor of pedicel cortical cells (as an indicator of pedicel water status) in situ. The in situ estimated hydraulic conductance of the water transport pathway between the vine and the berry decreased by about 90% from pre- to post-veraison. The hydraulic conductance at post-veraison was similar to the low conductance values reported in previous literature. Changes in pedicel cortical cell turgor were positively correlated with changes in SWP both at pre-veraison and post-veraison without a significant difference. This result indicates that the site responsible for the pre- to post-veraison decrease of berry hydraulic connection to the parent plant resided at a local scale, between the pedicel and the berry itself.
