Book Description
This book provides a broad and rich outline of the epigenetic mechanisms involved in cancer progression and the generation of metastasis. It describes the tumor suppressor genes undergoing transcriptional silencing by CpG island promoter hypermethylation in the different tumor types of the human anatomy and their association with tumoral behaviour. It also provides a comprehensive insightful look at the molecular players involved in DNA methylation, histone modification and chromatin remodelling complexes causing epigenetic lesions linked to the metastasic phenotypes. Finally, it explains how epigenetic lesions associated with cancer spreading can be targeted using new and potent chemotherapy drugs. The book is a state-of-the-art reference to all scientific researchers and clinicians interested in the understanding of the biological processes leading to tumor dissemination and to those that are keen to translate this knowledge to a better management of cancer patients. Each contributor is a specialist in their epigenetic area and their joint effort has created a unique view of the DNA methylation, histone and chromatin changes that define cancer metastasis.

Epigenetic Therapy of Cancer
Author: Michael Lübbert
Publisher: Springer Science & Business Media
ISBN: 3642384048
Category : Medical
Languages : en
Pages : 332
Book Description
The growing knowledge about disturbances of epigenetic gene regulation in hematopoietic stem cell disorders is now being translated into treatment approaches that target the epigenetic defects pharmacologically. This book first presents the latest evidence regarding the epigenetic regulation of hematopoietic stem cell differentiation and hemoglobin production. The significance of DNA methylation abnormalities in hematopoietic disorders and of epigenetic disturbances in lung cancer and other solid tumors is then discussed. A major part of the book, however, relates specifically to the translation of basic research and drug development to clinical applications, and in this context both present and future clinical strategies are considered. Individual chapters are devoted to the use of DNA hypomethylating agents and chromatin-modifying agents, and the treatment of hematologic malignancies and solid tumors by means of epigenetic agents is discussed in detail.

DNA Methylation Profiling Reveals Novel Biomarkers and Important Roles for DNA Methyltransferases in Prostate Cancer
Author: Yuya Kobayashi
Publisher: Stanford University

Category :
Languages : en
Pages : 76
Book Description
Candidate gene based studies have identified a handful of aberrant CpG DNA methylation events in prostate cancer (Brooks et al. 1998; Yegnasubramanian et al. 2004). However, large scale DNA methylation profiles have not been examined for normal prostates or prostate tumors. Additionally, the mechanisms behind these DNA methylation alterations are unknown. In this thesis, I describe the results of my efforts to better understand these previously unexplored areas of biology. For the study presented in this thesis, I quantitatively profiled 95 primary prostate tumors and 86 healthy prostate tissue samples for their DNA methylation levels at 26,333 CpGs representing 14,104 gene promoters by using the Illumina HumanMethylation27 platform. When the profiles of the prostate tissue samples were compared, I observed a substantial number of tumor-specific DNA methylation alterations. A 2-class Significance Analysis of this dataset revealed 5,912 CpG sites with increased DNA methylation and 2,151 CpG sites with decreased DNA methylation in tumors (FDR

Handbook of Nutrition, Diet, and Epigenetics
Author: Vinood B. Patel
Publisher: Springer
ISBN: 9783319555294
Category : Medical
Languages : en
Pages : 0
Book Description
This multivolume reference work addresses the fact that the well being of humankind is predicated not only on individuals receiving adequate nutrition but also on their genetic makeup. The work includes more than 100 chapters organized in the following major sections: Introduction and Overview; Epigenetics of Organs and Diseases in Relation to Diet and Nutrition; Detailed Processes in Epigenetics of Diet and Nutrition; Modulating Epigenetics with Diet and Nutrition; and Practical Techniques. While it is well known that genes may encode proteins responsible for structural and dynamic components, there is an increasing body of evidence to suggest that nutrition itself may alter the way in which genes are expressed via the process of epigenetics. This is where chemically imposed alteration in the DNA sequence occurs or where the functional expression of DNA is modulated. This may include changes in DNA methylation, non-coding RNA, chromatin, histone acetylation or methylation, and genomic imprinting. Knowledge regarding the number of dietary components that impact on epigenetic processes is increasing almost daily. Marshalling all the information on the complex relationships between diet, nutrition, and epigenetic processes is somewhat difficult due to the wide myriad of material. It is for this reason that the present work has been compiled.
