Biochemical Modulation at the present time defines an area of study in which the intracellular metabolism of a given anti cancer agent is modulated (usually by a noncytotoxic agent or a cytotoxic agent at sufficiently low dosage to make it non cytotoxic) in order to either increase the effectiveness of the particular agent against tumor cells or decrease its cytotox icity against normal cells. The major focus of modulation has been the agents 5-fluorouracil (FUra), arabinofuranosylcytosine (ara-C), methotrexate (MTX) and a few alkylating agents. The major thrust of the studies has been to increase the flow of the anticancer agent along the pathway responsible for the formation of the cytotoxic species: for example, FUra to FUTP or ara-C to ara-CTP. While in most cases the application of research re sults to clinical trials does not require the subsequent exper tise of the laboratory researchers, application of biochemical modulatory schemes to clinical protocols necessitate a dramatic break with the past procedures. As shown in the laboratory clinical loop below, close collaboration between the laboratory and clinical investigator is essential. While the laboratory REDEFINE TECHNOLOGY, TESTS OR QUESTIONS FOR FURTHER THERAPEUTIC ADVANCE CLINICAL EXPERIMENTAL PROTOCOL (LABORATORY) RESEARCH STUDIES DEFINE AND TEST APPROPRIATE SCIENTIFIC PARAMETERS results define rationally-based regimens, it is essential that the clinical protocols contain the requirement that clinical material (either tumor or normal tissues) be sampled to deter mine whether the biochemical modulation being proposed is in fact beinq accomplished.

The focus of the 22nd Annual Detroit Cancer Symposium was the presentation and discussion of cytotoxic agents, with a significant portion of the symposium including the exciting frontiers of drug discovery being explored by the National Cooperative Drug Discovery Groups (NCDDG) Program. The symposium brought together a large number of investigators from government, universities and pharmaceutical companies involved in the discovery and development of new anticancer agents. Exciting new leads were presented and the status of others presently under development was discussed. Of particular significance has been the initiation of renewed efforts in the area of natural product drug discovery, where the discovery of new cytotoxics is very promising at the moment. A number of major changes have occurred during the last decade in research on drug discovery of cytotoxic agents. Critical reviews of a number of the models and concepts underlying drug discovery represented a continuous thread throughout the meeting, being constantly discussed in terms of their advantages, disadvantages and capabilities of discovering solid tumor active anticancer agents. A recent development which is to be much applauded and which portends to great discoveries is the new relationship formed between Government, University of Industry. The NCDDG mechanism which stimulates this interaction is an inexpensive manner to greatly magnify the drug discovery and development effort nationally. This book is based on the invited and contributed papers presented at the 2nd International Conference on Anticarcinogenesis and Radiation Protection held at the National Bureau of Standards, Gaithersburg, Maryland, USA, on March 8-12, 1987. The conference documented developments that have taken place in areas that were addressed during the first conference in 1982. A number of new topics, such as biological response modifiers, were included because of their emerging relevance to anticarcinogenesis and radiation protection. The organization of the material in this book does not follow the conference program; rather, we have attempted to provide a different sequence for didactic reasons. The aim of the conference, which is reflected in this book, was to promote further development of mechanistic approaches to cancer prevention and treatment based on recent progress in molecular biology and free radical chemistry. At the basis of carcinogenesis lie changes in the dynamics of growth and differentiation of specific cell subpopulations in the target tissue. 'These changes are brought about by selective toxicity and modulation of gene expression that are induced by xenobiotic carcinogens and affected by physiological and genetic factors. This volume contains the proceedings of the First International Conference on Eicosanoids and Other Bioactive Lipids in Cancer and Radiation Injury held in Detroit, Michigan on October 11-14, 1989. The program consisted of 83 oral and 29 poster presentations, 74 of which are included in these proceedings. The major sponsors of the conference were the Armed Forces Radiobiology Research Institute, located in Bethesda, Maryland, the Radiation Oncology Research and Development Center of the Gershenson Radiation Oncology Center, Harper Hospital in Detroit, Michigan, and Schering AG of West Germany. Eighteen other organizations provided additional support. The conference was unique in its attempt to link the eicosanoid and lipid researchers in the radiobiology and cancer disciplines. The diverse roles that eicosanoids and other bioactive lipids play in these biological phenomena including the participation of lipid oxidation in conversion of procarcinogens, positive and negative modulation of tumor growth, immunomodulation, tissue injury, and yet protection and enhancement of cancer therapy, necessitated scientific interaction to sort out and understand these complex and sometimes contradictory observations. The success of this effort is reflected not only through these proceedings, but also through the decision to continue the conference series with a second meeting to be held in Berlin between September 17-21, 1991.

The European Organization for Cooperation in Cancer Prevention Studies (ECP) was established in 1981 to promote collaboration between scientists working in the various European countries on cancer causation and prevention. In order to achieve this aim, various working group- to deal with specific cancers or aspects of cancer aetiology, and to explore the opportunities for advances on a cooperative European basis - were established. It was also decided to hold annual symposia to draw general attention to fields in which there seemed to be many opportunities for progress in matters of prevention. These symposia have been devoted to themes of high priority to cancer prevention: "Tobacco and Cancer" (1983), "Hormones and Sexual Factors in Human Cancer Aetiology" (1984), "Diet and Human Carcinogenesis" (1985), "Concepts and Theories in Carcinogenesis" (1986)," Preventive Strategies for Cancer related to Immune Deficiencies" (1987), "Gastric Carcinogenesis" (1988), and "Breast, Ovarian and Endometrial Cancer: Aetiological and Epidemiological Relationships" (1989). This volume contains the proceedings of the 1990 ECP symposium held in Heidelberg, FRG, at the Deutsches Krebsforschungszentrum (DKFZ), on April 2-3 on "Causation and Prevention of Human Cancer".
