Essential Pathways and Circuits of Autism Pathogenesis
Author: Gül Dölen
Publisher: Frontiers Media SA
ISBN: 2889199053
Category : Genetics
Languages : en
Pages : 183
Book Description
The Centers for Disease Control and Prevention estimate that 1 in 68 children in the United states is afflicted with autism spectrum disorders (ASD), yet at this time, there is no cure for the disease. Autism is characterized by delays in the development of many basic skills, most notably the ability to socialize and adapt to novelty. The condition is typically identified in children around 3 years of age, however the high heritability of autism suggests that the disease process begins at conception. The identification of over 500 ASD risk genes, has enabled the molecular genetic dissection of the pathogenesis of the disease in model organisms such as mice. Despite the genetic heterogeneity of ASD etiology, converging evidence suggests that these disparate genetic lesions may result in the disruption of a limited number of key biochemical pathways or circuits. Classification of patients into groups by pathogenic rather than etiological categories, will likely aid future therapeutic development and clinical trials. In this set of papers, we explore the existing evidence supporting this view. Specifically, we focus on biochemical cascades such as mTOR and ERK signaling, the mRNA network bound by FMRP and UBE3A, dorsal and ventral striatal circuits, cerebellar circuits, hypothalamic projections, as well as prefrontal and anterior cingulate cortical circuits. Special attention will be given to studies that demonstrate the necessity and/or sufficiency of genetic disruptions (e.g. by molecular deletion and/or replacement) in these pathways and circuits for producing characteristic behavioral features of autism. Necessarily these papers will be heavily weighted towards basic mechanisms elucidated in animal models, but may also include investigations in patients.

Metabolism and Medicine
Author: Brian Fertig
Publisher: CRC Press
ISBN: 1000472140
Category : Medical
Languages : en
Pages : 453
Book Description
Chronic disease states of aging should be viewed through the prism of metabolism and biophysical processes at all levels of physiological organization present in the human body. This book connects these insights to what causes them to go awry in the context of unhealthy human behaviors and aging, aiming to buttress scientific creativity. It also provides links between the art and science of medicine that strengthens problem-solving in patient care. New and important discoveries in the area of metabolic health and metabolic diseases are discussed in exquisite detail. Key Features: Broad and up-to-date overview of the field of metabolic aspects of health and chronic disease development, especially connecting the spectrum of topics that range from molecular clocks to stress response to nuclear hormone receptors and the role of microbiota in human health Provides a deeper basic science and interdisciplinary understanding of biological systems that broaden the perspectives and therapeutic problem solving by elaborating on the usefulness of the Physiological Fitness Landscape Describes the importance of insulin resistance in metabolic disease, especially diabetes but also includes links to cancer and Alzheimer’s disease Examines the process of aging from the perspective of metabolic decline illustrating it with the Physiological Fitness Landscape This book, the second volume in a two-volume set, primarily targets an audience of clinical and science students, biomedical researchers and physicians who would benefit from understanding each other’s language.

Toward a Unified Ecology
Author: Timothy F. H. Allen
Publisher: Columbia University Press
ISBN: 0231538464
Category : Science
Languages : en
Pages : 505
Book Description
The first edition of Toward a Unified Ecology was ahead of its time. For the second edition, the authors present a new synthesis of their core ideas on evaluating communities, organisms, populations, biomes, models, and management. The book now places greater emphasis on post-normal critiques, cognizant of ever-present observer values in the system. The problem it addresses is how to work holistically on complex things that cannot be defined, and this book continues to build an approach to the problem of scaling in ecosystems. Provoked by complexity theory, the authors add a whole new chapter on the central role of narrative in science and how models improve them. The book takes data and modeling seriously, with a sophisticated philosophy of science.
