This is the third volume of the collection of new devices, modifications of existing equipment, and other items of interest of this nature published in the journal Applied Spectroscopy. These tricks have proved of value since they first appeared in the journal in 1959. They give solutions to many problems of workers in the var ious fields of spectroscopy. For the novice, the use of ali three vol umes may provide insight into the improvements that have been made in the instruments and techniques that he is currently using. The novice may be saved the necessity of discovering some shortcut that many experienced spectroscopists are already using. The contributions in this third volume are selected from the years 1970 through 1973. The subject arrangement is the same as in Volumes 1 and 2 according to the area of spectroscopy. Those tricks concerned with the same device are placed together so that the reader can easily compare them. To maintain the advantages in herent in a single collection of contributions, the subject index for this volume is cumulative including the tricks in the previous vol umes. Both author and journal indices are provided for this vol ume, the latter citing the original Applied Spectroscopy citation. The use of the contributions has been approved by the So ciety for Applied Spectroscopy, whose cooperation in this matter is gratefully acknowledged.

Spectroscopic Tricks was introduced in 1959 as a special section in the journal Applied Spectroscopy. Its purpose was to provide a means for communicating information on new devices, modifica tions of existing apparatuses, and other items of this nature of interest to the working spectroscopist. That it has proved valuable is indicated by the continuing publication of this section now under the title of Spectroscopic Techniques. However, the use fulness of these contributions, scattered through the many issues of the journal, diminishes as time passes since the reader must consult the annual indices of many volumes of the journal to find the contri bution that may hold the solution to his problem. The collection of the contributions into a single volume for the years 1959 through 1965 made it easier for the reader to make this search. The success of the first volume has prompted the continuation of these collec tions. The contributions in this second volume are selected from the the years 1966 through 1969. They are arranged in the same manner as in the previous volume according to the area of spectroscopy. Those concerned with the same devices are placed together so that the reader can compare them readily. To maintain the advantages inherent in a single collection of articles, the subject index for this volume includes all the entries and page references from the original volume. Both author and journal indices are are also provided, the latter citing the original Applied Spectroscopy edition.

Proteins are essential for all known forms of life and in many lethal diseases protein failure is the cause of the disease. To understand proteins and the processes they are involved in, it is valuable to know their structures as well as their dynamics and interactions. The structures may not be directly inspected because proteins are too small to be visible in a light microscope, which is why indirect methods such as nuclear magnetic resonance (NMR) spectroscopy have to be utilized. This method provides atomic information about the protein and, in contrast to other methods with similar resolution, the measurements are performed in solution resulting in more physiological conditions, enabling analysis of dynamics. Important dynamical processes are the ones on the millisecond timeframe, which may contribute to interactions of proteins and their catalysis of chemical reactions, both of significant value for the function of the proteins. To better understand proteins, not only do we need to study them, but also develop the methods we are using. This thesis presents four papers about improved NMR techniques as well as a fifth where the kinase domain of ephrinB receptor 2 (EphB2) has been studied regarding the importance of millisecond dynamics and interactions for the activation process. The first paper presents the software COMPASS, which combines statistics and the calculation power of a computer with the flexibility and experience of the user to facilitate and speed up the process of assigning NMR signals to the atoms in the protein. The computer program PINT has been developed for easier and faster evaluation of NMR experiments, such as those that evaluate protein dynamics. It is especially helpful for NMR signals that are difficult to distinguish, so called overlapped peaks, and the soft- ware also converts the detected signals to the indirectly measured physical quantities, such as relaxation rate constants, principal for dynamics. Next are two new versions of the Carr-Purcell-Maiboom-Gill (CPMG) dispersion pulse sequences, designed to measure millisecond dynamics in a way so that the signals are more separated than in standard experiments, to reduce problems with overlaps. To speed up the collection time of the data set, a subset is collected and the entire data set is then reconstructed, by multi-dimensional decomposition co-processing. Described in the thesis is also a way to produce suitably labeled proteins, to detect millisecond dynamics at C? positions in proteins, using the CPMG dispersion relaxation experiment at lower protein concentrations. Lastly, the kinase domain of EphB2 is shown to be more dynamic on the millisecond time scale as well as more prone to interact with itself in the active form than in the inactive one. This is important for the receptor function of the protein, when and how it mediates signals. To conclude, this work has extended the possibilities to study protein dynamics by NMR spectroscopy and contributed to increased understanding of the activation process of EphB2 and its signaling mechanism.

From the first appearance of the classic The Spectrum Analysis in 1885 to the present the field of emission spectroscopy has been evolving and changing. Over the last 20 to 30 years in particular there has been an explosion of new ideas and developments. Of late, the aura of glamour has supposedly been transferred to other techniques, but, nevertheless, it is estimated that 75% or more of the analyses done by the metal industry are accomplished by emission spectroscopy. Further, the excellent sensitivity of plasma sources has created a demand for this technique in such divergent areas as direct trace element analyses in polluted waters. Developments in the replication process and advances in the art of pro ducing ruled and holographic gratings as well as improvements in the materials from which these gratings are made have made excellent gratings available at reasonable prices. This availability and the development of plane grating mounts have contributed to the increasing popularity of grating spectrometers as com pared with the large prism spectrograph and concave grating mounts. Other areas of progress include new and improved methods for excitation, the use of controlled atmospheres and the extension of spectrometry into the vacuum region, the widespread application of the techniques for analysis of nonmetals in metals, the increasing use of polychrometers with concave or echelle gratings and improved readout systems for better reading of spectrographic plates and more efficient data handling.
