The goal of this project was to explore the sensory mechanisms which control expression of bioluminescence in the marine bacterium Vibrio harveyi. Genetic methods were used to discover a complex network of genes which encode functions for the production of extracellular, chemical signals (autoinducers) and for the synthesis of signal receptors (sensor kinases and response regulators). The genes and proteins defined by this study resemble elements of the phosphorelay paradigm known as two-component signal transduction. Therefore, the Vibrio harveyi system is considerably different from the quorum sensing mechanism used by other luminous bacteria.

Genetic methods were used to discover the genes which encode functions for the production of extracellular, chemical signals (autoinducers) and for the synthesis of proteins which mediate the response to such signals. A mutant defective in the production of one class of autoinducers was isolated. The gene or genes defective in this mutant were not isolated, but this mutant should be useful for future work to identify specific signaling functions. Another gene, luxT, which is important for transcription of the luxT genes encoding the luminescence enzymes, was cloned and sequenced. Further work to understand its precise role must still be done.

Bioluminescence is everywhere on earth—most of all in the ocean, from angler fish in the depths to the flashing of dinoflagellates at the surface. Here, Thérèse Wilson and Woody Hastings explore the natural history, evolution, and biochemistry of the diverse array of organisms that emit light. While some bacteria, mushrooms, and invertebrates, as well as fish, are bioluminescent, other vertebrates and plants are not. The sporadic distribution and paucity of luminous forms calls for explanation, as does the fact that unrelated groups evolved completely different biochemical pathways to luminescence. The authors explore the hypothesis that many different luciferase systems arose in the early evolution of life because of their ability to remove oxygen, which was toxic to life when it first appeared on earth. As oxygen became abundant and bioluminescence was no longer adequate for oxygen removal, other antioxidant mechanisms evolved and most luminous species became extinct. Those light-emitting species that avoided extinction evolved uses with survival value for the light itself. Today’s luminous organisms use bioluminescence for defense from predators, for their own predatory purposes, or for communication in sexual courtship. Bioluminescence was earlier viewed as a fascinating feature of the living world, but one whose study seemed unlikely to contribute in any practical way. Today, bioluminescence is no longer an esoteric area of research. Applications are numerous, ranging from the rapid detection of microbial contamination in beef and water, to finding the location of cancer cells, to working out circuitry in the brain.

Bacterial reporters are live, genetically engineered cells with promising application in bioanalytics. They contain genetic circuitry to produce a cellular sensing element, which detects the target compound and relays the detection to specific synthesis of so-called reporter proteins (the presence or activity of which is easy to quantify). Bioassays with bacterial reporters are a useful complement to chemical analytics because they measure biological responses rather than total chemical concentrations. Simple bacterial reporter assays may also replace more costly chemical methods as a first line sample analysis technique. Recent promising developments integrate bacterial reporter cells with microsystems to produce bacterial biosensors. This lecture presents an in-depth treatment of the synthetic biological design principles of bacterial reporters, the engineering of which started as simple recombinant DNA puzzles, but has now become a more rational approach of choosing and combining sensing, controlling and reporting DNA 'parts'.
