Ce travail présente dans un premier chapitre des généralités sur les comprimés, les principales données à connaître sur la compression directe à travers ses avantages et inconvénients, l’importance des excipients pour compression directe et les caractéristiques du mélange à sec. Il définit également la validation de façon générale et en particulier la validation de procédé utilisée dans ce transfert de production. Faisant suite à ces principes théoriques, le deuxième chapitre expose les divers travaux réalisés par l’étude des formules et des matières premières utilisées, sur des procédés de mélange à sec et pour la création de toute la documentation de validation requise (dossiers de fabrication, protocoles et rapport de validation). Le dernier chapitre décrit les résultats obtenus sur les lots d’essai et de validation lors de la fabrication des différentes formules. Ainsi, les résultats de la validation ont permis de mettre à jour les dossiers d’A.M.M. pour déclarer, d’un point de vue réglementaire, le nouveau site comme « site de fabrication » et modifier certaines normes de fabrication.

Pour remédier à l'empirisme qui domine le plus souvent la formulation des comprimés, de nombreux auteurs ont utilisé les techniques modernes de mesure physique dans le domaine de la compression : ces chaînes de mesures équipant les machines permettent entre autre la mesure de FORCES au niveau des organes de compression (poinçons inférieur et supérieur, matrice, levier d'éjection) et du DEPLACEMENT du poinçon supérieur. Depuis, de très nombreux travaux tous très originaux ont été réalisés et ceci tant au niveau recherche-développement qu'au niveau des machines de production. Et bien que déjà très exploité, ce domaine de la physique de compression cache encore beaucoup d'informations. A travers notre travail, nous avons voulu d'abord présenter d'un point de vue très général la technique de mesure à l'aide des chaînes instrumentées: pour ce faire, nous avons d'abord présenté l'appareillage nécessaire, pour ensuite montrer quelques-unes des possibilités d'informations apportées par ce système de mesures et enfin terminer sur un exemple d'application au niveau développement galénique. Dans une seconde partie (expérimentale) cette fois, nous avons mis en application les données précédemment acquises : connaissant le matériel, nous avons mis en place un protocole de validation du système de mesure ; sur ce matériel fiable, nous avons appliqué la méthode de mesure à un exemple concret en réalisant une étude comparative des paracétamols (qui a confirmé les résultats des tests empiriques) ; cette dernière a permis de déboucher sur une formulation de comprimés de paracétamol par compression directe. Bien qu'encore au stade d'étude (étant donné le temps imparti) cette dernière étape nous a permis de mettre en application une "méthodologie de formulation" qui prend de plus en plus le pas sur l'empirisme.
