Premature Chromosome Condensation: Application in Basic, Clinical, and Mutation Research emerged from a workshop supported by the International Union against Cancer that reviewed the status of research on premature chromosome condensation and explored future possibilities, not least in relation to cancer. The workshop was held at the Institut für Humangenetik, Freie Universität Berlin, on September 6, 1980. The book begins with a review of the process of premature chromosome condensation. This is followed by separate chapters on the functional and morphological aspects of premature chromosome condensation; the behavior of prematurely condensed chromosomes (PCC) during mitosis in binucleate and multinucleate cells; chromosomes from spermatogenic cells; and micronucleus-derived PCC. Subsequent chapters discuss the application of banding techniques to the study of PCC; the factors involved in the condensation of interphase chromatin into chromosomes; and the induction of premature chromosome condensation in somatic plant cells. This book will be helpful to readers working in basic and applied research or in teaching. This year we remember the 39th anniversary of the atomic bomb explo sions in Hiroshima and Nagasaki, which led to the exposure of thou sands of people to high doses of ionizing radiations. Nearly 18 years earlier, on the 15th of September, 1927, H. J. Muller presented his paper The Problem of Genic Modification at the Fifth International Congress of Genetics in Berlin, in which he brilliantly demonstrated the muta genic activity of X-rays. In 1928, K. H. Bauer formulated his mutation theory of the origin of cancer, and already in 1914, Th. Boveri speculat ed that tumor cells originate from an abnormal chromosomal comple ment. In the meantime we have learned that also nonionizing radiation and an immense number of environmental chemicals, both, man-made and naturally occurring, are mutagenic in a variety of test systems, in cluding human cells. In no case has it been shown unequivocally that physical or chemical mutagens have led to an elevation of the mutation rate in the germ cells of man, but in view of the huge body of experi mental data this seems to be a problem of detection. It can be expect ed that germ cell mutations are induced as a consequence of exposure to mutagens in man, as yet undetectable with the methods at hand. An uncontrolled addition of mutations to the human gene pool may well have unforeseen and catastrophic consequences in future genera tions for whom we should feel responsible.

Since the book was first written, human genetics has undergone dramatic developments, mainly due to the introduction of new concepts and techniques from molecular biology. Con comitantly, "basic" scientists have become increasingly interested in problems of human genetics. More than 700 human genes have been mapped, genes of previ ously unsuspected complexity -such as the gene for factor VIII - have become known, and the structure of noncoding DNA sequences is being analyzed with the aim of understanding gene regulation. DNA diagnosis is being rapidly intro duced into medical genetics. All this, as well as the extensive progress in most other fields of human and medical genetics, had to be considered in the prepara tion of this second edition. The book has been extensively revised and rewritten. A substantial new section dealing with gene and chromosomal structure at the molecular level has been added. The newer knowledge of molecular genetics has been incorporated, and the conceptual and practical contribution of DNA methods (for example in the hemoglobinopathies and in some other diseases) is discussed.
