Volume II provides a comprehensive and detailed source of information concerning the genetics and regulation of biological nitrogen fixation in free-living prokaryotes. This preface attempts to provide the reader with some insight into how this volume originated, how it was planned, and then how it developed over the several years of its production. Once the editorial team was established, the first job was to decide which of the many free-living diazotrophs that have been subjected to genetic analysis should be included in this volume. Would we need to develop specific criteria for selection or would the organisms, in effect, select themselves? Of course, Klebsiella pneumoniae and Azotobacter vinelandii, which have served (and still serve) as the main model organisms for the genetic analysis of diazotrophy, plus some of the other bacteria described in this volume, did indeed select themselves. However, there was considerable discussion surrounding well-characterized fixing species, like Azorhizobium caulinodans and Herbaspirillum seropedicae, both of which are able to fix atmospheric N under free-living conditions.

The turn of the millennium from the twentieth to the twenty-first century provides an occasion to review our understanding of a biological process, biological nitrogen fixation, that is of prime importance for the continued survival of mankind. This process has provided a basis for maintaining soil fertility since the beginning of organised agriculture, yet its very existence was confirmed only just over a century ago. In the intervening years, an enormous intellectual effort has dispersed much of the mystery surrounding biological nitrogen fixation. Biological fixation is widely exploited in agriculture, as are nitrogen fertilisers prepared for the last hundred years under extreme conditions of temperature and pressure. An Fe- and Cu-Protein Symposium was held on December 15-18, 1975 at the East West Center-University of Hawaii and was sponsored by the United States-Japan Cooperative Science Program under the auspices of the National Science Foundation and the Japan Society for the Promotion of Science. It was recognized by the organizers of the symposium that metalloproteins are very important in the field of health science and a subject worthy of discussion by experts from the United States, Japan and Europe. The meeting was restricted to Fe- and Cu-proteins but this is still a very broad subject matter and therefore, selected topics of current interest in this field were chosen. This book contains the collected papers from most of the symposium participants. The subject matter covered in this book is divided into four parts. These are: 1) the iron-sulfur proteins (which are not a part of the mitochondrial electron transport system); 2) the iron-sulfur proteins and heme proteins of the mitochondrial electron transport system; 3) other heme and nonheme iron proteins; and 5) selected copper proteins. The organizers of the symposium wish to express their gratitude to the participants, the session chairmen, and Drs. I.C. Gunsalus and E. Frieden who assisted in the organization of the symposium.

Nitrogen and sulfur compounds are continuously synthetized, degraded and converted into other forms in nature. There are many similarities in the principle problems and basic mechanisms of the biology of inorganic nitrogen and sulfur. Many details are not yet understood and hence are the subject of active investigation the world over. In May, 1980, a conference was held in Bochum, Federal Republic of Germany, at which attempts were made to discuss and compare all aspects of both the nitrogen and the sulfur cycle. Lectures were given by internationally recognized experts on the physiology, biochemistry, genetics, and ecology of dinitrogen fIXation, of assimilatory and dissimilatory nitrate and sulfate reduction, and of ammonia and sulfide oxidation. In addition, important data were communicated by German scientists of the national program on the Metabolism of Inorganic Nitrogen and Sulfur Compounds, supported by the Deutsche Forschungsgemeinschaft. This book contains all the contributions to the meeting and consequently should be of interest to researchers, teachers, and students in the field.
